This study was conducted to investigate the effect of phytohormone concentration in the tissue culture medium on the mass propagation of Syngonium podophyllum 'Pixie' and the effect of growing medium condition on the acclimation of plantlets after in vitro propagation. In primary culture for 16 weeks, superior adventitious bud and shoots for subculture were obtained when plant tissues were cultured on MS medium with 1.0 mg/L of 6-benzylamino purine (BA). To do mass proliferation of those plants, the shoot tips of Syngonium podophyllum 'pixie' were cultured on MS medium with BA concentrations of 0, 0.5, 1.0, 3.0 and 5.0 mg/L and thidiazuron (TDZ) concentrations of 0, 0.05, 0.1, 0.5 and 1.0 mg/L, respectively. The formation of adventitious bud and shoot propagation were affected by phytohormone concentrations. The formation of adventitious buds and shoot length increased with increasing BA concentration from 0 to 3.0 mg/L. However, there was no effect from a further increase in BA concentration. The effect of TDZ on the formation of adventitious bud was similar to that of BA. The shoot length was decreased significantly when shoot tips were cultured with 1.0 mg/L of TDZ. Maximum number of shoots was obtained when shoot tips were cultured with 3.0 mg/L of BA and 0.5 mg/L of TDZ. To evaluate the effects of IBA and NAA concentration on shoots growth and rooting in vitro of Syngonium podophyllum 'Pixie', the sections of shoot were cultured for 4 weeks on the MS medium containing 0.5 g/L of charcoal with IBA and NAA concentrations of 0.5, 1.0, 3.0 and 5.0 mg/L. Regardless of IBA and NAA concentrations, all plants rooted. Shoot length, root length and number of roots increased with increasing IBA concentration from 0.5 to 3.0 mg/L. However, there was no effect from a further increase in IBA concentration. Root length increased with increasing NAA concentration from 0.5 to 5.0 mg/L, while the NAA concentrations had no effect on the shoot length and number of roots. When plants were treated with higher concentration of IBA and NAA(5.0 mg/L), the roots were turned to dark brown color and decomposed. These seedlings were not marketable. To evaluate the effects of growing medium condition on acclimation of in vitro cultured plantlets, we grew plantlets in three different growing media (peat moss versusperlite ratio; 3:7, 5:5 and 7:3, v/v). Plantlets were transplanted into 5 cm round pots filled with growing medium. All plants were placed on benches in a greenhouse. The temperature set points for the greenhouse were 30 ℃ during the day and 25 ℃ at night. More than 98% of plantlets survived, regardless of the growing medium. At 4 weeks after transplanting, plant height, number of roots and root length increased at higher peat moss ratio versus perlite ratios (5:5 and 7:3, v/v) compared to the 3:7 peat moss versus perlite ratio.
