카포시육종 헤르페스바이러스 (Kapoi's Sarcoma-associated Herpesvirus, KSHV) 또는 사람헤르페스바이러스-8(Human Herpesvirus-8, HHV-8)은 사람 혈관내막세포 또는 B 세포에 감염하여 카포시육종(Kaposi's sarcoma, KS), 또는 PEL, MCD 등의 세포과증식증후군이나 종양을 유발하는 감마헤르페스바이러스의 하나이다. HIV 감염 등으로 인하여 면역기능이 저하된 환자에서 KSHV 감염으로 인하여 유발되는 카포시육종은 후천면역결핍증후군 증상 중 주요 증상의 하나로 피부 및 여러 실질장기를 침범하는 종양질환으로 HHV-8이 원인 병원체임은 밝혀져 있으나, 명확한 발병기전 또는 종양화 기전에 대해서는 실험모델의 부재로 인하여 밝혀진 바가 거의 없다. 바이러스감염으로 인한 종양화 기전 연구는 바이러스 감염에 따른 숙주세포 유전자 발현의 변화를 먼저 확인하고, 과다 발현 또는 과소발현되는 유전자를 선별하여 그 유전자가 가지는 생물학적 의미를 고찰함으로써 해석될 수 있다. 이에 본 연구자는 기존에 수립된 카포시육종연관바이러스?사람혈관내막세포 감염 모델을 이용하여 감염 후 발생하는 사람혈관내막세포 유전자 발현의 변화를 유전자칩을 이용하여 관찰하고, 분석하였으며, 유의하게 변화를 보이는 유전자 중 세포의 생존과 종양 발생에 관여하는 것으로 생각되는 세포주기 조절 유전자, 세포자멸사 관련 유전자, 손상된 유전자 복구와 관련된 유전자를 대상으로 감염 시기에 따른 발현량을 분석하였으며, 바이러스감염이 이들 유전자의 발현을 조절함으로써 성공적인 바이러스 증식을 이루어냄과 동시에 감염된 세포의 누적된 손상과, 손상 복구의 차단이 바이러스에 의한 종양발생의 중요한 기전이 될 수 있음을 밝혔다.

Kaposi’s Sarcoma-Associated Herpesvrus (KSHV), also known as Human herpesvirus 8 (HHV8) plays a significant role in the development of Kaposi’s sarcoma (KS), primary effusion lymphoma (PEL) and some forms of multicentric Castleman’s disease (MCD). Study on the pathogenesis of HHV8 had been hampered by the lack of reliable infection model before the KSHV?BAC36 - HUVEC infection system had established. Using KSHV-BAC36 - HUVEC infection system, we tested the modulation of cellular gene expressions by infection of KSHV with microarray technique. Many functionally related gene expressions were altred in time-course manners. In here we tested several categories of genes that related to cell cycle management, apoptosis management, and DNA repair system. The KSHV infection on HUVECs were significantly altered cell cycle and the cell grows rapidly, blocked apoptosis, and hampered the functions of DNA repair system. The orchestration of those gene expression modulation could be a main cause of tumorigenesis of Kaposi’s sarcoma.
