A survey of sweetpotato virus disease was carried out in nursery bed as field, and virus infection rate was 5~100% and 100% at sweetpotato nursery bed and field, respectively. The incidence and severity of the diseases were shown not to have any relationship in area and cultivars grown. A total of 179 samples were collected and analyzed based on serological, electron microscopic and molecular properties. Field-grown sweetpotatoes were examined to inspect 8 different viruses using NCM-ELISA, resulting that 30% of sweetpotato was infected by one virus and 70% was by more than 2 viruses. However, RT-PCR using primers designed and selected for eight viruses, such as sweetpotato feathery mottle virus(SPFMV) revealed that 40% of the 179 samples were infected with SPFMV, SPGV(sweetpotato virus G) with 16%, mixed infection of both viruses with 11% and 27% had unknown viruses. Especially, SPGV(sweetpotato G virus) and sweetpotato leaf curl virus(SPLCV) were first detected, whereas not sweetpotato chlorotic stunt virus(SPCSV) in the sweetpotato growing areas in Korea. In root, viral diseases were severer in Yeoju than in Mokpo Experiment Station and infection rate was much different depending on sweetpotato cultivars. A total of 56 samples for SPFMV(sweetpotato feathery mottle virus) and SPGV(sweetpotato G virus) were subjected to restriction fragment length polymorphism (RFLP) analysis, using of coat protein region primers with 7 enzymes to assess genetic variation. A 1.186bp-CP (coat protein) encoding fragment for SPFMV and 1.277kb for SPGV was amplified by RT-PCR. Sequence variation among samples was characterized by RFLP analysis and confirmed by sequencing. By digestion of CP-encoding region with 7 enzymes each, SPFMV was classified into three groups, whereas SPGV into two groups. Phylogenetic analysis of the aligned coat protein clustered SPFMV group Ⅰ in the 97% SPFMV-O(Japan) subgroup, and SPFMV group Ⅱ and Ⅲ? in the SPFMV-RC(U.S.A) subgroup. The sequence of SPGV showed 97%, 96% and 86% identity with SPGV-Eg(Egypt) and SPGV-CH, and CH2(China), respectively. Virus-free plantlets of Yulmi, Shinjami and Sinhwangmi in-vitro cultured, were grown out in insect-proof greenhouse. The total plant weight and weight of the aboveground part were examined in order to determine the most appropriate nutrient solutions, culture media and cutting intervals. The results showed that growth increment of plantlets, in all cultivars, was high in nutrient solutions of 0.5 to 1.0 times higher than the Horticulture of Standard Nutrient Solutions, provided that the plantlet were planted in medium B with a cutting interval of 20 days. Likewise, in order to improve the efficiency of cutting production in plug trays in insect-proof greenhouse, single-node and double-node leafless cuttings were planted on 72-plug tray and 128-plug tray and the growth increment was investigated. According to the result, plants cultivated in 72-plug tray showed longer branch length and vigorous growth than those on 128-plug tray. And to determine the most appropriate culture media, tray size, and nutrient solutions for cuttings, two-node plants were planted, and then the plant length, the number of nodes, diameter and fresh weight were examined. The result showed that media B was superior to vermiculite and that 72-plug tray was better than 128 plug one. For this purpose, virus free plants were planted on rockwool+perlite, loess, and sand media to investigate the yield of tuberous roots. Compared to rockwool+perlite or loess media, sand media showed a yield increased of 39~59% in Yulmi, 68% in Shinjami, and 21~40% in Sinhwangmi. The infection rate of the aboveground part in harvesting season indicated a 0% infection. In open-field cultivation, two methods were used: insect-proof(net) and non-insect proof. Though the yield of virus-free plants propagated in the non-insect proof open-field cultivation was 2,402kg/10a, 6% higher than those in the insect-proof cultivation, but the rate of virus re-infection was 18% higher compared to 3.3% with insect-proof cultivation.
