The purpose of this study was to isolate of AMPV and define the subtypes of AMPV isolates in Korea by ELISA, subtype specific RT-PCRs, and cell culture. Serum samples were tested for AMPV antibodies usging commercial enzyme linked immunosorbent assay (ELISA) kit. AMPV antibodies were detected in 513 out of 579 chicken sera (88.6 %), and were found in chickens of all ages, ranging from 1 days to over 60 weeks. The trachea and tracheal swabs from 30 domestic chicken farms inoculated onto chicken embryo allantoic cavity. Viral RNA from allantoic fluid were analyzed by subtype specific RT-PCR. The results, 5 samples (1 out of 65 layer samples, 4 out of 23 brolier samples) of 88 samples were RT-PCR positive for AMPV RNA. The results of RT-PCR based on subtype specific primers indicated subtype C AMPV were present in Korea. And Newcastle disease virus (NDV), Infectious bronchitis virus (IBV) were also isolated from the AMPV positive samples. AMPV positive samples were continued until passage five, inoculated into vero cell and chicken embryo fibroblast (CEF) cell. The isolates replicated in cell culutre without CPE, including syncytia, patches of dead and sloughing cells. However, the presence of AMPV in vero cell and CEF cell was confirmed by RT-PCR. In this investigation, we confirmed avian metapneumovirus infection in chickens in Korea, and expected that the virus and other agents could induce economic losses. In conclusion, the Korea poultry industry, vaccination programs should be adjusted to include the AMPV vaccine to aid in the control of this respiratory disease.
