Hydroxyapatite (HA) and porous ceramics have been widely investigated for application to bone scaffolds to improve biocompatibility, bone bonding ability, and bone regeneration. Ideal scaffolds for bone regeneration requires highly interconnected porous structure with porosity of 50∼90 % and pore size of at least 100 ㎛ for bone ingrowth and vascula rization, along with bioactivity. HA with such a porous structure is so weak in strength (ie., 0.5∼7.4 MPa) that its application has been restricted to non-load bearing implants such as bone augmentation and extenders. On the other hand, Al₂O₃ has superior mechanical properties so that relatively higher strength is expected with the highly porous structure. If porous Al₂O₃ which is bioinert is coated with bioactive HA, it could be used suitably as load bearing bone substitutes.
In this study, porous HA and HA coated porous Al₂O₃ possessing the pore characteristics required for bone substitutes were prepared by a slurry foaming method combined with gelcasting. This method is advantageous in producing porous ceramics with a cellular structure with high degrees of shape complexity and control of pore size and connectivity. The HA coating was deposited simply by heating porous Al₂O₃ substrates in an aqueous solution containing Ca^(2+) and PO₄^(3-) ions at 65∼95 ℃ under the ambient pressure. Coated samples were also prepared with disc substrates to compare coating behavior and measure bonding strength of the coating. The density, pore properties, microstructure, and compressive strength were investigated and compared for the porous HA and Al₂O₃ samples. And the microst ructure, thickness, crystallinity and bonding strength of the coating layer were investigated for the HA coated samples.
The density and porosity of the porous samples were about 0.75 g/㎤, 80 % for Al₂O₃ and 0.62 g/㎤, 81 % for HA with a highly interconnected network of spherical pores with size ranging from 50 to 300 ㎛. The porous Al₂O₃ sample showed much higher compressive strength (25 MPa) than the porous HA sample (10 MPa).
Fairly dense and uniform HA coatings were deposited on the porous Al₂O₃ with fine rod-like crystals. The bonding strength of the coating was found to be 8 MPa. The coating thickness increased with the repetition of coating process, resulting in about 2 ㎛ for the porous substrate and 4.5 ㎛ for the disc substrate with three times deposition.
Since the compressive strength of cancellous bone 2∼12 MPa, both the porous HA and HA coated Al₂O₃ samples could be successfully utilized as bone scaffolds substituting for cancellous bone. Especially the HA-coated Al₂O₃ samples is expected to showed a good performance in load bearing parts due to its excellent strength.
