Apigenin is a plant flavonoid and a pharmacologically active agent that has been isolated from several plant species. However, the molecular mechanism of apigenin mediated-immune modulation is not fully understood. One of the possible mechanisms for its protective properties is by downregulation of the inflammatory responses. In the present study, the human mast cell line (HMC-1) was used to investigate this effect. Apigenin significantly inhibited the induction of inflammatory cytokine such as tumor necrosis factor (TNF)-α, interleukin (IL)-8, IL-6 and granulocyte-macrophage colony-stimulating factor (GM-CSF) by phorbol 12-myristate 13-acetate (PMA) plus A23187. Moreover, apigenin attenuated cyclooxygenase (COX)-2 expression and intracellular Ca^(2+) levels. Apigenin inhibited PMA plus A23187-induced nuclear factor (NF)-κB activation, IκB degradation. Furthermore, apigenin suppressed the expression of TNF-α, IL-8, IL-6, GM-CSF, and COX-2 through a decrease in the intracellular Ca^(2+) levels, and also showed a suppression of the IκBα degradation and NF-κB activation. These results indicated that apigenin exerted a regulatory effect on inflammatory reactions mediated by mast cells.
