In many recent previous studies, cockroaches have been suggested as an important component of house dust allergens and having an important role in atopic disease, In Korea, two most common cockroach species are German cockroach (GE, Blatella germanica) and the American cockroach (AE, Periplaneta americana). In spite of recent advances in the purification and characterization of cockroach allergens, identification of clinically important allergens and their source have not been completely elucidated. This present study was undertaken to investigate a group of atopic patients to specific extracts of American and German cockroaches to asses the prevalence and importance of cockroach allergens by SDS-PAGE and Western blotting techniques. The results are summarized as follows: 1. In SDS-PAGE analysis of both cockroach extracts, AE revealed a lot of allergenic bands with a MW 5 to 110 kd. The pronounced major bands at between 92-55, 50, 45, 36-38, 28-32, 14-20, 5 kd with many minor staining protein. GE revealed a lot of allergenic bands with a MW 5 to 110 kd. The pronounced proteins at 98-64, 50, 45, 24-18, 16, 5 kd. 2. In SDS-PAGE - separated extract proteins immunoblotted with a pooled serum sample from cockroach-allergic subjects, AE shows IgE-binding to proteins at 67, 64, 55, 36, 30 kd. GE shows IgE-binding to proteins at 92, 67, 64, 55, 30, 18, 5 kd. 3. In SDS-PAGE-separated extract proteins immunoblotted with a 9 serum sample from cockroach-allergic subjects, a major common allergen of AE, found in 9 of 9 sera tested, was a protein band in 55 kd. In GE, 55 kd was found in 9, 64 kd in 8 of sera tested. Other bands in GE exhibiting significant activity were the 50 kd band with 78%, the 30 kd band with 45%, the 67 kd band with 33%, the 92kd band with 22%. In conclusion, four allergens (MW 67, 64, 55, 30 kd) can be considered as a probable shared allergen among the two cockroach extracts. And, in Korea, the 55 kd allergen is the only major allergen in AE, compared to 64 & 55 kd allergen in GE. Our study also indicates that there may be specific allergens that include bands in AE of MW 36 kd, a band in GE of MW 92, 16, and 5 kd.
