AI-2 생산 억제능력을 비교한 결과 B. longum ATCC 15707과 B. adolescentis ATCC 15706균주는 control에 비해서 각각 약 92-fold, 67-fold 의 억제능력이 관찰되었다. 또한 biofilm 억제능력을 비교한 결과 B. longum ATCC 15707과 B. adolescentis ATCC 15706 균주는 control에 비해서 각각 약 36%와 32%의 biofilm 생성 억제능력이 관찰되었다. 따라서 본 연구에서는 E. coli O157:H7 ATCC 43894의 AI-2 생산 및 biofilm 생성 모두에서 억제 능력이 가장 우수한 것으로 관찰된 B. longum ATCC 15707균주를 최종 새로운 개념의 선택적 병원성 미생물 저해 물질 생산 균주로 선발하였다. B. longum ATCC 15707에서 추출한 세포질 성분에 대한 E. coli O157:H7 ATCC 43894의 AI-2 생산성과 biofilm 생성 억제작용을 조사한 결과, 20 mg/mL의 세포질 추출물을 첨가한 경우 AI-2 생산 억제능력이 가장 높게 나타났으며 E. coli O157:H7의 성장에는 영향을 주지 않았다. 세포질 성분에 protease K (10 μg/mL)를 처리하였을 때 AI-2 생산 억제능 및 Biofilm 생성 억제능력이 확연하게 감소되는 것으로 보아 이 물질은 단백질성 물질임을 확인 할 수 있었다. B. longum ATCC 15707과 B. adolescentis ATCC 15706 세포질 추출물을 E. coli O157:H7 ATCC 43894에 처리 시 AI-2 저해 효과를 단백질 발현변화로 확인하기 위하여 이차원 전기영동을 수행하였다. Nonlinear IPG strip (pH 3~10)을 이용하여 등전점 전기영동 후 12.5% SDS-PAGE에서 이차원 전기영동을 실시한 다음 spot을 관찰한 결과 각 gel의 image에서 평균적으로 250개의 단백질 발현을 확인할 수 있었으며 PDQuest image analysis software (Bio-Rad, USA)를 이용하여 최소 2.5배 이상의 발현 차이를 보이는 7개의 spot을 선발하였다. 7개의 spot을 대상으로 Matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF/MS)와 proteome database를 이용하여 단백질 규명 및 관련 기능을 확인한 결과 철의 대사 또는 생합성과 관련된 물질대사를 조절하는 단백질을 coding하는 nifU, 운동성과 세포막 단백질과 관계있는 유전자인 flgI, 산화를 촉진 시키거나 독성요소들이 발현될 때 필요로 하는 유전자 dsbA의 발현은 억제 되었다. 그러나 hybE의 발현은 증가하는 것으로 나타났다. 결론적으로 B. longum ATCC 15707의 세포질 추출물에 존재하는 단백질성 물질이 AI-2 유사활성을 억제시키고 biofilm 생성을 억제하는 것으로 확인되었으며, 이물질은 E. coli O157:H7 ATCC 43894에 있어서 metabolic process, cell motility, secretion과 envelope biogenesis에 관련된 유전자의 발현을 억제시키는 것으로 관찰되었다.

This study was investigated to determine the effect of Bifidobacterium sp. on the quorum-sensing (QS) signals and biofilm formation of E. coli O157:H7 ATCC 43894. Inhibition of QS signals measured by autoinducer-2 (AI-2) activity was investigated in the presence or absence of cell-free supernatants and cell extracts of Bifidobacterium sp. The AI-2 activity was measured for E. coli O157:H7 ATCC 43894 using the V. harveyi coupled bioluminescent assay. Comparison of AI-2 inhibition by the different Bifidobacterium sp. showed a wide variation with a range from 0 to 92-fold. Bifidobacterium longum ATCC 15707 exhibited the highest inhibitory activity (92-fold inhibition) for all the Bifidobacterium strains tested. In an AI-2 bioassay, the addition of 10 mg/mL of B. longum ATCC 15707 cell extracts to supernatants of E. coli O157:H7 resulted in 98-fold decrease in AI-2 activity in E. coli O157:H7 as well as in the V. harveyi reporter strain. However, it did not inhibit the growth of E. coli O157:H7. In a biofilm bioassay, the addition of 10 mg/mL of B. longum ATCC 15707 cell extracts to supernatants of E. coli O157:H7 resulted in 36% decrease in biofilm formation. As expected, the E. coli O157:H7 exhibited high AI-2 activity. When compared to control, the biofilm formation increased when they were exposed to AI-2 activity. This shows that AI-2-like activity plays a key role in the pathogenicity of E. coli O157:H7. Susceptibility of inhibitory substance to a protease was performed by incubating with protease K (10 μg/mL) at 30°C for 4 hr. After incubation, the enzyme were inactivated by heat treatment at 60°C for 10min and tested for AI-2 activity. Inhibitory substance was found to be sensitive to protease K. This result indicates that the inhibitory substance is proteinaceous in nature. Furthermore, the proteomic analysis was investigated the role of AI-2 on the E. coli O157:H7 cellular proteins using a two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF/MS) analysis. The protein expression patterns in the presence of 20 mg/mL cell extracts isolated from B. longum ATCC 15707 and B. adolescentis ATCC 15706 were studied. Seven proteins were differentially expressed in the E. coli O157:H7 when supplemented with cell extracts of B. longum ATCC 15707 and B. adolescentis ATCC 15706. 2-DE analysis revealed that Bifidobacterium cell extracts up-regulated two proteins and down-regulated five proteins. Among these protein spots, formation of iron-sulfur protein (NifU), thiol:disulfide interchange protein (DsbA), flagellar P-ring protein (FlgI), hydrogenase-2 operon protein (HybE) were identified by MALDI-TOF/MS. The FlgI protein which is involved in motility was down-regulated in the presence of Bifidobacterium cell extracts suggesting the involvement of signaling moelcules other than AI-2 on motility of E. coli O157:H7. These identified proteins may associate with metabolic process, adaptation and protection, cell motility, secretion, envelope biogenesis, and protein translation. The results of this study support the need for further evaluation of potential QS inhibitors of Bifidobacterium sp. for effects on toxin production and biofilm formation in food matrix.
