In recent years, it has become increasingly apparent that posttranscriptional control of gene expression in eukaryotes is important in various cellular processes such as growth, development, and stress adaptation. Contrary to the increasing amount of knowledge regarding the functional roles of glycine-rich RNA-binding proteins (GRPs) and zinc finger-containing GRPs (RZs) in Arabidopsis thaliana in stress Es, the physiological functions of GRPs and RZs in rice (Oryza sativa) currently remain largely unknown. In this thesis, the functional roles of the gene family encoding OsGRPs and OsRZ from rice were investigated under cold or freezing stress conditions. Among the six OsGRPs and three OsRZs investigated, OsGRP1 (Os01g68790), OsGRP4 (Os04g33810), OsGRP6 (Os12g31800), and OsRZ2 (Os07g08960) were shown to have the ability to complement cold-sensitive BX04 E. coli mutant cells under low temperature conditions, and this complementation ability was correlated closely with their DNA- and RNA-melting abilities. Moreover, OsGRP4 and OsRZ2 complemented the cold-sensitive phenotypes of grp7 mutant plants, in which the expression of AtGRP7, one of the eight GRPs in Arabidopsis, was suppressed and is sensitive to cold and freezing stresses. Considering that AtGRP7 confers freezing tolerance in plants and harbors RNA chaperone activity during the cold adaptation process in E. coli, the results of the present study provide evidence that GRPs in rice and Arabidopsis are functionally conserved, and also suggest that GRPs perform a function as RNA chaperones during the cold adaptation processes in monocotyledonous plants, as well as in dicotyledonous plants. In this thesis, the expression and functional roles of four microRNAs including miR395c, miR395e, miR402, and miR414 were investigated in Arabidopsis plants subjected to a variety of stress conditions. The miRNAs investigated in this thesis have been predicted to regulate the expression of several target mRNAs; miR395c and 395e for ATP sulfurylase and sulfate transporter, miR402 for DNA glycosylase (DEMETER) and ATP-dependant helicase, and miR414 for diverse groups of mRNAs. The transgenic Arabidopsis plants that constitutively overexpress miR395c or miR395e survived better than the wild-type plants under sulfate starvation conditions. Overexpression of miR395c retarded the germination of Arabidopsis plants under high salt or dehydration stress conditions, whereas overexpression of miR395e accelerated the germination of the plants under the same stress conditions. Considering that miR395c and miR395e differs each other at only one nucleotide position, these results demonstrate that a single nucleotide difference in a particular family of miRNA can guide the cleavage of different mRNA targets and perform a function as a positive or negative regulator of seed germination in Arabidopsis plants under stress conditions. Analysis of the transgenic Arabidopsis plants showed that miR414 negatively affects the seed germination of the plants under salt stress and ABA treatment conditions, while miR402 positively affects the seed germination and seedling growth of the plants under salt or dehydration stress and ABA treatment conditions. The results of the present study provide a basis for further investigation of the regulatory roles of miRNAs in the responses of plants to a variety of stress conditions.
