Due to its strong demand in pharmaceutical area resulting from irreplaceable role in hematopoiesis, the price is tremendously high. In this study, we tried to produce hEPO from transgenic chickens by injection of concentrated retrovirus solution beneath the blastoderm of freshly laid eggs (stage X). Because it is generally believed that constitutive over-expression of some cytokine genes in the transgenic animals may cause serious physiological disturbances, the vectors were designed to express the hEPO gene in the presence of tetracycline. When the G0 transgenic chicken was fed with doxycycline (0.5 mg per 1 gram of feed), a tetracycline derivative, hEPO concentration of the transgenic chicken blood was 350 IU/㎖. As we expected, the same chicken without doxycycline treatment showed almost no hEPO expression. No evident physiological disturbances have been observed and we demonstrated successful germline transmission of the transgene by generating G1 progeny from a normal hen mated with a transgenic rooster. The biological activity of the recombinant hEPO was comparable to its commercially derived CHO cell counterpart. The resulting tetracycline inducible vector system may be helpful in solving serious physiological disturbance problems which have been a major obstacle in successful production of transgenic animals.
