The present study was designed to investigate the effect of shoots of Acanthopanax senticosus against water immersion restraint (WIR)-induced stress and forced swimming-induced fatigue. The hot-water fraction of the plant was studied on the ulcer index, adrenal gland mass and biochemical changes in stressed rats. Also, it was estimated that effect of the extracts on swimming time in mice using adjustable current swimming pool apparatus. To evaluate anti-stress effect of the extracts, male SD rats were fed with shoots of Acanthopanax senticosus 100 mg/kg/day, p.o. for a week. End of the pretreatment day, rats were sacrificed immediately after a 4 hour WIR stress, the blood was collected, adrenals and spleen were dissected for organ weight and stomach for ulcer score. WIR stress exposure significantly increased the ulcer index, bleeding score, adrenal gland weight, plasma glucose, total cholesterol, cortisol, and creatine kinase but sigmificantly decreased the spleen weight. In addition, WIR stress significantly increased TBARS and myeloperoxidase level, while significantly decreased GSH level and antioxidant enzymes such as superoxide dismutase, catalase, glutathione peroxidase in stomach. These oxidative damages and hyper activation of the HPA axis were reversed effectively by pre-treatment of the extracts. To determine anti-fatigue activity of the extracts, the swimming capacity of male ICR mice was studied with an adjustable-current swimming pool. Total swimming period until fatigue was measured as the index of the swimming capacity. Mice were fed shoots of Acanthopanax senticosus 1 g/kg body weight, p.o. daily until end of the experimental period. Swimming time was measured every other day for 10 days. The swimming endurance capacity of mice orally administered with the extract was significantly prolonged. The swimming-induced fatigue showed significant body weight loss, and administration of the extracts limited a decrease of body weight. Since they inhibited hyper activation of HPA axis and oxidative damages induced by WIR stress and prolonged the swimming time, it can be considered that the extract of shoots of Acanthopanax senticosus has anti-stress and anti-fatigue effects.
