3. 항아토피 효과에서 혈중 Ig E 수준과 비장세포 배양 상층 액에서 IL-5와 IL-13 생산량 수준, ALN과 등 피부 조직의 총세포수, PBMCs에서 CD11b+/Gr-1+ 세포, ALN에서 CD4+ Th 세포, B220+/CD23+ 세포, 등 피부조직에서 CD4+ Th 세포, CD11b+/Gr-1+ 세포의 총 절대 세포 수, 그리고 등 피부조직에서의 IL-4, IL-13, COX-2, TNF-αmRNA 유전자 발현은 산조인 복합오일(ZjJ-C_oil), 독활 복합오일(ARC-C_oil)에서 대조군에 비하여 통계학적으로 유의성 있게 감소되었다. 실험 종료 후 등 피부를 절단하여 H&E 염색 하여 분석한 결과, 정상군에 비하여 대조군(CT)의 상피조직이 부종으로 현저하게 확장되었고 leucocytes의 침윤, erythema, edema, excoriation, scaling이 보이고 있었으며, 산조인 복합오일(ZjJ-C_oil), 독활 복합오일(ARC-C_oil)에서 대조군(CT)에 비하여 상피조직의 두께가 현저하게 줄었고, leucocytes의 침윤 및 erythema, edema, excoriation, scaling 등은 보이지 않아 부종이 거의 사라진 것을 알 수 있다. 또한 등피부에 침윤된 비만세포를 관찰하기 위하여 toluidine blue 염색 하여 분석한 결과, 정상군에 비하여 대조군(CT)의 비만세포가 현저하게 증가되었고, 산조인 복합오일(ZjJ-C_oil), 독활 복합오일(ARC-C_oil)은 대조군(CT)에 비하여 비만세포가 거의 사라진 것을 알 수 있었다. 결론적으로, 산조인 복합오일(ZjJ-C_oil), 독활 복합오일(ARC-C_oil)은 아토피 접촉 피부염 유발 NC/Nga mouse에서 면역학적 염증 반응을 조절하는 데에 유의적인 효과가 있다고 볼 수 있으며 이러한 기전을 바탕으로 아토피 피부염에 대체 치료법으로 이용 가능할 것으로 판단된다. 또한 독활은 외부에서 주어지는 통증에 대한 민감도 개선에는 효과가 없지만, 통증 개선에는 효과가 있는 것으로 사료되고, 산조인은 항스트레스에 효과가 있는 것으로 판단된다.

This study aims to investigate the anti-atopic, anti-nociceptive and anti-stress effects of essential extracts from Zizyphus jujuba var. spinosa and Aralia cordata var. continentalis. For this purpose, animal experiments were carried out. In order to verify the anti-atopic efficacy, DNCB(1-chloro 2,4-dinitrobenzene) was applied to NC/Nga mice to induce atopic dermatitis, and then the effects of Zizyphus jujuba var. spinosa Composite oil(ZjJ-C_oil) and Aralia cordata var. continentalis Composite oil(ARC-C_oil) on the suppression of skin rash symptoms of atopic dermatitis were tested. An injection of onosodium iodoacetate(arthritis-inducing agent) into the one leg of the male SD rat was administered to induce a localized arthritis, and after the repeated oral administration of Aralia cordata var. continentalis, the weight bearing index and paw withdrawal threshoulds were measured to evaluate the pain relief effects. To verify the anti-stress efficacy, the open field test(OFT) and elevated plus maze test(EPM) were conducted after the repeated inhalation of natural essential oil mixtures in the male SD rat. The results are as follows.
1. In the anti-stress effect, the test substance-treated group (50% dose) showed a statistically significant decrease compared with the negative control group in the freezing item. There was a statistically significant difference between the test substance-treated group(100% dose) and the negative control group in the locomotion activity, mean speed, rearing and freezing items. The measurement of the adrenal weight revealed that the negative control group did not show a statistically significant difference from the normal control group, but there was an increase in the negative control group. In conclusion, the natural essential oil mixture, which is a test substance in the male SD rat, has an anti-stress effect at the dose level of 100%.

In conclusion, both the ZjJ-C_oil and the ARC-C_oil have significant effects on the inhibition of immunological inflammatory reactions in atopic contact dermatitis-induced NC/Nga mice, and thus can be used as alternative treatments for atopic dermatitis.
