Site-specific DNA methylation과 hTERT 발현과의 상관관계 분석은 hTERT의 높은 발현을 보이는 HCT116(대장암세포주), MCF7(유방암세포주)의 hTERT-고발현군과 상대적으로 낮은 발현을 보이는 HeLa(자궁경부암세포주), SK-OV3(난소암세포주)의 hTERT-저발현군에서 또한 수행되었는데 그 결과 hTERT-고발현군에서는 SP1 단백질의 결합부위인 14^(th), 15^(th), 22^(nd) CpGs가 demethylation되어 있고 CTCF 단백질의 결합부위인 34-36^(th) CpGs가 강하게 methylation 되어 있는 반면 hTERT-저발현군에서는 반대의 양상을 보이는 것으로 관찰되었다. 또한 각 세포주에서의 SP1과 CTCF의 결합은 해당 결합부위의 CpG methylation에 의해 강하게 조절 받는 것으로 관찰되었다. 두 그룹의 세포주에서 hTERT의 발현은 histone deacetylase의 억제제인 TSA처리에 의해 서로 다르게 조절되었는데, TSA는 hTERT-고발현군에서는 그 발현을 감소시키는 반면 hTERT-저발현군에서는 증가시키는 것으로 관찰되었다. 따라서 TSA에 의한 hTERT의 서로 다른 조절기작을 분석하기 위해 hTERT전사조절 부위의 DNA methylation양상이 분석되었는데 그 결과 고발현군에서는 TSA처리후 hTERT 전사조절부위의 DNA demethylation이 유도되는 반면 저발현군에서는 DNA methylation 양상의 변화가 없음을 관찰 할 수 있었다. 이러한 DNA methylation의 서로 다른 변화는 두 그룹에서의 DNA me^(th)yltransferase인 DNMT1의 발현 양상의 변화와 일치하였는데 고발현군에서는 TSA에 의해 DNMT1의 발현감소가 관찰된 반면 저발현군에서는 DNMT1 발현의 변화가 관찰되지 않았다. 또한 DNMT1의 발현조절 기작 분석을 위해 TSA에 의한 p21과 p53의 발현변화를 살펴본 결과 고발현군에서는p53과 p21의 증가가 관찰된 반면 불활성화된 p53을 갖는 저발현군에서는 p21의 증가를 관찰 할 수 없었다. 즉 두 그룹에서의 TSA에 의한 DNMT1의 발현억제는 세포내 기능적인 p53의 존재유무에 따른 p21의 발현 유도와 관련되어 있으며 이러한 일련의 결과들이hTERT의 site-specific methylation 상태를 변화시켜 hTERT 발현을 억제함을 확인할 수 있었다. 본 연구결과는site-specific CpG methylation과 관련된 hTERT의 후생학적 발현조절 기작을 규명 함으로서 향후 다양한 암세포에서 hTERT의 발현 조절을 응용한 치료에 대한 접근을 용이하게 할 것이다.

hTERT, which encodes the catalytic subunit of telomerase and is expressed in most immortalized and cancer cells, has been reported to have increased DNA methylation pattern on the promoter for the expression. This pattern is inconsistent with general observation that the DNA methyaltion on the promoter is typically associated with gene silencing. In this study, to elucidate the more detail epigenetic mechanism of hTERT expression by DNA methylation, we performed the hTERT expression assay and methylation analysis of the site-specific CpGs on the promoter of hTERT in colorectal carcinoma tissues and various cancer cell lines showing various clinical stage and differential expression level of endogenous hTERT respectively. As a result, we observed an increased pattern of hTERT expression according to the malignant progression of colorectal carcinoma. In particular, the expression of hTERT began at Duke's B stage of colorectal carcinoma and significantly increased at the turning point from Duke's B to Duke's C. The increase of hTERT expression was induced by site-specific methylation of the 7^(th), 2^(nd)~10^(th) and 11^(th) CpGs including putative binding sites of SP1 and AP2. In this study, it was found the cancer cell lines of two groups showing differential expression level of the endogenous hTERT. HCT116 (Colon cancer cell line) and MCF7 (Breast cancer cell line) were identified the high expressed group of the hTERT (ht-High) whereas HeLa (Cervical cancer cell line) and SK-OV3 (Ovarian cancer cell line) were identified the low expressed group of the hTERT (ht-Low). To investigate what caused the different expression of hTERT in the two groups, we performed the analysis of the site-specific CpG methylation in the cancer cell lines. The ht-High showed completely demethylated pattern on the 14^(th), 15^(th) and 22^(nd) CpGs of Sp1 binding sites and fully methylated pattern on the 34-36^(th) CpGs of CTCF binding sites but, on the contrary, ht-Low showed the partial methylated pattern on the CpGs of SP1 and CTCF binding sites. Chromatin immunoprecipitation (ChIP) analysis reveals that SP1 bound to the hTERT promoter in both groups. However, CTCF bound to the hTERT promoter in the ht-High but not in ht-Low. It means that the binding of SP1 and the releasing of CTCF by site-specific methylation was required for enhanced expression of hTERT in cancer cells. Interestingly, two groups showed the differential regulation of hTERT expression in response to TSA (Trichostatin A; histone deacetylase inhibitor) treatment. The hTERT expression after TSA treatment was significantly decreased in the hT-high but slightly increased in the ht-low. We also observed that TSA induced the demethylation of the CpGs on the hTERT promoter and down-regulation of DNMT1 (DNA methyltransferase 1) in hT-high but not in hT-low. It suggests that the repression of hTERT in ht-High was caused by the demethylation of the site-specific CpG on the hTERT promoter via down-regulation of DNMT1 after TSA treatment. Recently, it was known that DNMT1 repress p21 transcription and p21 negatively regulates DNMT1 expression in a feedback mechanism. In our results, the expression of p21 was significantly increased in ht-High but not in ht-low after TSA treatment. The increasing was dependent on the induction of p53 in ht-High, which has intact p53. However, the expression of p21 in ht-low was not observed and it might be caused by the dysfunction of endogenous p53. Additionally, induction of p53 in ht-high inhibits the binding of SP1, transcriptional activator for the hTERT expression, on the hTERT promoter. In conclusion, TSA repressed the expression of hTERT via demethylation of the CpGs on the hTERT promoter and inhibition of SP1 binding on the hTERT promoter by p53. The demethylation of the CpGs was mediated by the down-regulation of DNMT1 according to the induction of p21 and p53 after treatment with TSA. These finding suggests that hTERT expression is epigenetically regulated by site-specific CpG methylation and provides an important clues for approaching cancer-specific therapies using epigenetic regulation of hTERT expression in various cancer cell types.
