중간엽 줄기세포는 동일한 암 비글견의 골수•지방•진피 로부터 외과적으로 회수되었고, 10% 소 혈청이 함유된 ADMEM을 기본배양액으로 하여 5% CO2, 100% humidity, 37.5ºC 인큐베이터 속에서 배양 되었다. 그리고 각 세포들의 배양액은 3일 간격으로 교체 되었다. 이 실험을 위하여 사용된 세포의 일령은 6 passages 미만을 사용하였다. 실험 결과 골수•지방•진피 유래 중간엽 줄기세포들 모두 outgrowth로 인하여 colonies를 성공적으로 형성하였다. AP 활성은 진피 유래의 중간엽 줄기세포에서 가장 강하게 발현됐고, 골수 유래는 약간 덜 발현되었으며, 지방유래에서는 약하게 발현되었다. 저농도 의존적 CFU-F 분석 결과, colony의 크기는 골수, 지방, 그리고 진피 순으로 유의적으로(P<0.05) 증가하였고, colony 수는 지방 골수 피부 순으로 유의적으로(P<0.05) 감소 하였다. 초기 줄기세포 전사 인자들인 Oct3/4•Sox2•Nanog는 세가지 조직원 중 지방유래 중간엽 줄기세포가 골수와 진피 유래 보다 강하게 발현됨을 관찰할 수 있었다. 중간엽 줄기세포 특이 CD marks중에서 negative marker인 CD45는 진피와 골수 유래 둘 다에서 발현되지 않았지만 지방유래에서는 약하게 발현됨이 관찰되었다. 중간엽 줄기세포의 positive marker인 CD90은 지방 골수 피부유래 중간엽 줄기세포 모두에서 발현됨을 확인 할 수 있었고 특히 지방에서 강하게 발현 됨을 확인할 수 있었다. 세포주기분석 결과에서는 지방 골수 피부유래 중간엽줄기세포들 모두에서 비교적 많은 S phase로 인하여 비교적 적은 G0/G1 phase가 관찰 되었다. 염색체 배수성분석 결과 지방 골수 피부유래 중간엽 줄기 세포들 모두가 90% 이상의 정상적인 핵형(Diploid)을 관찰 할 수 이었다. 지방 골수 피부유래 중간엽 줄기세포간에 telomere의 길이 분석결과 세가지 중간엽 줄기세포 모두 21kbase 전후로 유의적인(P<0.05) 차이를 관찰 할 수는 없었지만, 지방유래 줄기세포가 다소 telomere의 길이가 긴 경향을 보였다. 다분화능 확인을 위하여 지방 혹은 골세포로 3주 동안 분화 유도 결과 지방 골수 피부유래 중간엽 줄기 세포들 모두 지방세포와 골세포로 분화가 되었고, 특히 지방 유래 중간엽줄기세포가 Oil red o 와 Alizalin red S로 염색한 결과 가장 강하게 발현 됨을 확인 할 수 있었다.
이상의 결과들을 통합하여 볼때 지방 골수 피부유래 중간엽 줄기세포들 중 지방 유래 중간엽 줄기세포가 증식능, 중간엽줄기세포 특이 CD marks의 발현능, Telomere 길이, 체외에서 안정적인 분화능을 보임으로 가장 잠재력 있는 중간엽 줄기세포원임을 확인할 수 있었다. 따라서 이러한 연구결과는 개와 사람의 치료적 적용을 발전시키고 표준화하는데 기초자료로 활용될 수 있을 뿐만 아니라 대단위 전 임상 실험을 통한 tissue engineering과 재생의학적 관점에서 중간엽 줄기세포의 치료적 역할 규명에 활용될 수 있을 것이다.

Adult stem cells that can be found in a wide range of adult tissues are an item of infinitely ascending interests because of their biological properties, and are identified as potential tools for cell therapy, regenerative medicine and tissue repair. In addition, MSCs have been under intense attentions due to their ability to self-renew and differentiate into multi-lineages in canine and human. Particularly, canine MSCs have proven as pivotal models for overcoming neurological diseases and modulating immune properties. Furthermore similarities between human and canine MSCs documented by several studies allow canine MSCs to be considered as a valuable model system for in vitro studies and cell based therapy.
A few studies have been reported the effective isolation and expansion of canine MSCs isolated from different origins such as bone marrow, fat tissues, dermal skin and umbilical cord blood. In canine, the identity of MSC still remains uncertain as there is no single universally accepted immunological definition and little standardization in the procedures used to isolate and culture such cells. In addition, there are no quantitative assays to assess the presence of MSCs in any given population.
The present study was therefore designed to establish animal model using MSCs and composed of two parts; Part 1, Establishment of culture conditions of MSCs derived from different origins and Part 2, Characterizations of MSCs derived from different origins, with following objectives; 1) to develop isolation and culture condition of MSCs derived from different origins by evaluating alkaline phosphatase (AP) activity and colony formation and 2) to evaluate the characterizations of MSCs derived from different origins by investigating pluripotential ability (Oct3/4, Sox2 and Nanog), the expressions of specific CD markers (CD45, 90 and 105), cell cycle, ploidy, differentiation ability to adipocytes and oseteocytes, and telomere length. MSCs were collected from dermal skin tissue, fat tissues and bone marrow in the same animals.
MSCs were successfully isolated and cultured under modified conditions from different origins; dermal skin tissues, fat tissues and bone marrows, and had formed colonies in those from all origins. AP activity in MSCs isolated from dermal skin tissues was intensively expressed, and followed by bone marrows. However, in fat tissue, feeble expression was observed. By observation of CFU-F assay, the size of colonies in MSCs from bone marrow appeared bigger than that from fat tissues, and the number of colonies in MSCs derived from fat tissues had significantly higher than that from bone marrow. However, no colony was formed in MSCs from dermal skin. MSCs isolated from fat tissues, were strongly expressed pluripotential markers, Oct3/4, Sox2, Nanog compared to those from dermal skin tissues and bone marrows. CD45, as a negative marker in MSCs, were not expressed both in dermal skin tissues and bone marrows. Interestingly, the weak expression revealed in fat tissues, causing to unclear reason. CD90, as a positive marker in MSCs, were expressed in all of three different sources, but the intensity of expression was higher in fat tissues. CD105, as a positive marker in MSCs, were higher expressed in fat tissues than that in bone marrows, but did not expressed in dermal skin tissues. Higher incidence of S phase of the cell cycle revealed in all of three different sources, however, G0/G1 phase was lower. This characteristic was remarkable in dermal skin tissues. Most of MSCs isolated from different sources have diploid chromosomal sets, but in fat tissues observation of tetraploidy was significantly higher that other two sources. No differences in the length of telomere were appeared in all of three different sources, but bone marrows tended to be slightly shorten the length. MSCs isolated from three different sources were successfully differentiated into adipocytes and oesteocytes, respectively, at 3 weeks of culture in induced medium.
However, non-availability of universally accepted markers selective for canine MSCs has proven difficult to standardize the cell culture, even CD45, 90 and 105 could be adopted in the present study. The differentiation of canine MSCs derived from different origins into mesenchymal lineages, adipocytes and oesteocytes, under in vitro conditions is consistent and demonstrated a considerable level of plasticity.
Collectively, MSCs isolated from fat tissues have higher stem cell potency than other sources, and hence they could be adapted for the cell therapy as an alternative, although those from three different origins have similarity of stem cells characterizations. Furthermore, successful isolation, characterization and in vitro differentiation of canine MSCs into multi-lineages along with other findings provide additional support in developing and standardizing therapeutic strategies for canine and human applications. In additions, easy obtaining and purification of MSCs from dermal skin are regarded them as ideal candidates.
Preclinical assessments in a large animal scale would intensely facilitate the realization of a major therapeutic role for MSCs in tissue engineering and regenerative medicine, and give rise to highly relevance to humans. To assure the probable short and long term safety and efficacy of MSC therapy, adequate preclinical trials need to be performed for the next study.
