Background and Objectives: The purpose of this study is to clarify the role of epinephrine, stress-related hormone, on hair cells of normal cochlea. Materials and Methods: Adult ICR mouse was divided into two groups; the control group was treated with intratympanic 0.9% normal saline while the experimental group was treated with 1:10,000(epinephrine with concentration of 91μg/ml, 0.1ml injection) epinephrine. ABR threshold and pathologic changes were detected in both groups. Also, toxicity and signal changes following epinephrine application in HEI-OC1 cells were measured using MTT assay and calcium imaging. Results: ABR threshold increased to maximal point 10 minutes after intratympanic 1:10,000 epinephrine injections which returned to normal range. Also, there was marked elongation and folding of Reissner’s membrane after epinephrine applications. But there were no observable changes of cell viability in MTT assay after epinephrine administration and there were no alterations in F340/F380 fluorescence ratios before and after the epinephrine injection. Conclusion: Epinephrine did not have direct toxicity on hair cells neither exhibit signal changes related to calcium. But the morphologic changes like elongation and folding of Reissner’s membrane developed after topical injection of epinephrine with reversible ABR threshold alterations. The stress-related hormone epinephrine influences the endolymphatic fluid component but further mechanism is left to be solved in the future.
