Traumatic brain injury (TBI) is a complicated disorder which is associated with social development and induces severe damage or deadly outcome. It causes the multiple staged-procedure of injury including primary mechanical impact and its related secondary damage. It is also involved neuroprotective cellular and molecular responses, however, this beneficial effect is limited and transient. Thus, the therapeutic strategy is needed to maintain or promote the endogenous brain recovery mechanism. The purpose of the present study is to investigate the effects of forced treadmill and spontaneous wheel running exercise on brain recovery following traumatic brain injury in rats. To confirm the impact of TBI and effect of running exercises, 60 Sprague-Dawley male rats were used and divided randomly 6 groups (NRM, TBI1, TBI2, TBI3, FTR, and SWR group). TBI was induced by using modified cortical controlled impact model. Moreover, neurological behavior tests, Triphenyltetrazolium chloride staining, immunohistochemistry, western blot analysis, RT-PCR, and the others were measured. In the present study, TBI increased morphological damage and apoptotic protein expression. Moreover, neurotrophic factors such as BDNF, NGF, and NT-3 were decreased significantly after transient increase by TBI. Applications of treadmill and wheel running following TBI induced the significant improvement in neurological behavior functions, morphological recovery, neurogenesis as well as the expression of apoptotic proteins, neurotrophic factors, stress-induced proteins, synaptic plasticity proteins, and Ca2+-dependent proteins. Especially, treadmill running induced more increase than wheel running in the expression of stress-induced proteins and synaptic plasticity proteins. In conclusion, TBI induced neurological and histological damage in brain tissue, whereas treadmill and wheel running exercise following TBI induced the improvement of neurological behavior functions and brain recovery-related proteins.
