Research Background: Humane sensory organs receive and process various kinds of external stimulations. Among these, tactile sensation is a direct response to external stimuli and causes different kinds of emotional and cognitive reactions. Recent nervous imagery studies show the signals from tactile stimulation are processed by complex cerebral nerve network. Especially the feelings such as tickling sensation not only activate emotion-related area but also the area related to cognitive sensations. However, those questions as whether tickling sensation has its own nervous system distinctive to others or exactly what relationships exist with cognito-emotional functions such as laugh for example, haven't answered yet. And therefore, in this study, through functional magnetic resonance imageries, an attemp is made to clarify the cerebral network responsible for tickling sensation, which is distinctive from the rest of other networks processing normal stimulations. Moreover, beyond the sensory information process, we examined the relationship to laughing mechanism which is closely related to tickling. For this, functional magnetic resonance images are used see the differences in activated area of cerebral cortex under normal and tickling sensations for both healthy male and female. Subject and method: 16 healthy adults(Average age: 28.9/ Right handed: 14, Left handed: 2) took places in this research and the experiment was processed after a sufficient explanation and consents by the participants. Before the imagery experiment, a pre-survey was given to select suitable tools and body parts for tickling stimulation. Scraggy scrubbers were used for normal sensation and swabs for tickling sensation. The stimulated areas for the experiment were sole of the foot. This imagery experiment included 2 steps of image acquisitions, 3DT1 for finding anatomical reference point and EPI for detecting functional changes in cerebral cortex under stimulations with time duration of 5 minutes 35 seconds and 4 minutes 5 seconds respectively and 16 minutes and 2 seconds in total, including survey and reference scanning time. During the functional image acquisition, stimulations were suggested according to reversely-balanced block designs and one identical person gave the stimulations for objectivity and consistency in stimulation suggestion. Acquired images went through image pre-treatment and statistical analysis using SPM99. Individual analysis for each subject and group analysis depends on group condition were conducted and by using one sampled t-test, the relationship between the activated area and normal and tickling stimulation is examined. The activated area showing differences in those two conditions were examined for any statistical significance using pair-sampled t-test. In additions, the correlation analysis was carried out during the group analysis to see the range in which shows significant correlations to percepted tickling strength. The whole analyzing processes for testing significant correlations with parameters of uncorrected t=0.001 and voxel size = 20. Result: Individual analysis showed the activation of somatic sensation area in both tasks and the tickling sensation test showed more activated area in the Wernicke's area(BA40) compared to the normal sensation. The group analysis result shows that under normal stimulations, both sides of somatosensory cortices(BA 1, 2 and 3) were activated and under tickling stimulation, not only the cortices but also those huge activation on thalamus, cingulate gyrus and insular lobe were detected and when the tickling was stopped, significant activations were shown in right cingulate gyrus, left MFG area and left insular lobe. Moreover, the correlation analysis show a significant activation on right frontal lobe, left cingulate gyrus and right MFG area. Conclusion: Following the pre-research result, both the normal and tickling stimulations showed a significant cerebral activation in somatosensory cortices(BA 1, 2 and 3). In ticklling condition, activation of the insular cortex, which is responsible for laughing, sadness or happiness and the medial prefrontal cortex responsible higher recognition and social activities was shown. The activation of insular cortex, which wasn't activated under the similar stimulations, suggests that tickling process isn't just a tactile responses however, more related to the emotional aspects. In addition, the laughing-related area, which has an important role in socio-mutual activities was also activated. The result shows that the primitive stimulation such as tickling is much closely related to laugh, which is an important factor for various social activities. Through this study, a cerebral area responsible for recognizing tickling sensation is examined and thus, further comparisons on cerebral activation mechanism of different stimulations such as visual or acoustic are expected.
