Confocal Laser microscopy를 이용한 IFA 에서 recombinant protein (PvCDPK4-EF)가 mature schizont stage에서 특이적으로 발현함이 관찰되었으며, 이는 Pf-PDPK1과 유사한 반응으로 말라리아 원충생활환의 증식 또는 egress 과정에 관여한다고 사료된다. 이러한 Ca2+ signaling에 관여하는 kinases 들의 지속적인 연구를 통해 stage specific한 anti-malarial drug targets으로써의 유용성이 기대되었다. 2. 국내 삼일열 말라리아 antigenic markers의 유전학적 다양성 조사 국내 삼일열 말라리아는 재유행 후 지속적인 발생을 통하여 점차 토착화되고 있는 가운데 유전자 다형성이 다양해지면서 위험지역도 남쪽지역으로 확산되고 있다. 국내에 발생되는 삼일열 말라리아와 해외유입말라리아의 관찰 및 재발/재감염의 감시를 위하여 국내 발생 말라리아에 대한 molecular genotyping 및 분자역학분석을 통한 데이터베이스의 구축은 매우 중요하다. 본 연구에서는 삼일열 말라리아 연구의 유용한 markers로써, 높은 polymorphism을 보이는 4가지 항원유전자 (merozoite surface proteins-1 (PvMSP-1), circumsporozoite proteins (PvCSP), apical membrane antigens-1 (PvAMA-1), 그리고 duffy binding protein (PvDBP))를 이용하여 최근 국내에서 발생하는 삼일열 말라리아 환자들의 유전학적 다형성을 조사하고, 지역 및 연도별 특징을 관찰하였다. 최근 국내에서 발생한 156명의 환자혈액을 이용하여 원충 유전자에 대한 PCR을 실행하고 이 유전자를 분석하였다. 결과, MSP-1은 6가지, AMA-1과 DBP는 4가지 타입의 아형이 확인되었다. 또한 CSP VK210에서는 repeat motif 의 반복에 대한 5가지의 haplotypes으로 나뉘었다. 본 연구에서 새로운 타입의 AMA-1 과 DBP의 아형을 발견하였다. 또한, 156개 시료의 4가지 유전자 subtype combination set를 분석하여 최근 국내 삼일열 환자들의 dominant한 유전자 타입 set를 도출하고, 적어도 60개의 combination set를 확인하여 유전자 다형성이 증가하고 있음을 확인하였다. 국내에서 발생되고 있는 유전자의 분자역학분석 (4가지 항원유전자 조합 분석)을 바탕으로, 19명의 재발/재감염 환자의 유전자 분석을 통하여 그 원인을 분석해냈다. 또한 국내발생으로 보고된 환자를 유전자 분석을 통하여 해외유입 환자로 구별하였고, 잠복기의 시기에 대한 연구가 수행되었다. 즉, 4가지 유전자 (MSP-1, CSP, AMA-1그리고 DBP)는 국내 삼일열 말라리아 환자의 유전자 분석을 위해marker 로써 이용될 수 있으며, 이러한 protocol은 재발/재감염환자와 해외유입환자를 구별해 냄으로써 낮은 비용으로 빠르고 정확한 genotyping data를 구축할 수 있는 유전학적 tool을 제공할 것으로 사료된다.

Plasmodium vivax is becoming increasingly resistant to many currently available drugs. Increased efforts to identify novel targets for anti-malaria therapy are required and some protein kinases and/or the corresponding genes have been isolated from several parasites. One of these, Calcium dependant protein kinases (CDPK) found in plant, green algae, ciliates and apicomplexa parasite, including malarial parasites, but not in mammals. In Plasmodium, members of the CDPK family are candidates for key regulators of stage-specific cellular responses to Ca2+, which is controlling the progression of the parasite through its complex life cycles. Recent studies have demonstrated that CDPK implicated in a number of steps involving motility and development in Plasmodium. According to CDPK genotypes of P. falciparum, it shows composition of four distinct domains, which have Ca2+ binding regions to calmodulin-like domain. This finding support the hypothesis that P. vivax may have similar one. This is the first report on CDPK protein characterization in P. vivax. In this study, identified PvCDPK4 sequence consists of 1,761 bp and encodes 545 amino acids with a predicted molecular mass of approximately 62 kDa. This gene has several domains including calmodulin-like domain and contains four EF hand Ca2+ -binding motifs, which suggest that Ca2+ activates PvCDPK4 directly. Then it focused that cloning and expression regions of calmodulin-like domain and reported molecular characterization. The recombinant protein (rPvCDPK4-EF) has molecular weight about 34 kDa and shows a positive immune reaction with serum from vivax patients in South Korea. The rPvCDPK4-EF expressed at the mature schizont stage of parasite. The expression of rPvCDPK4 in schizont may suggest that it participates in the proliferation or egress process in parasite’s life cycle. These protein kinases could represent potential anti-malarial drug targets, leading to the development of drugs more specific to the parasite and also act as a possible.part II
Molecular genotyping is required for better understanding of the molecular epidemiology of Plasmodium vivax and will help to predict and monitor the effectiveness of intervention strategies, such as the success of therapeutic regiments, the spread of drug resistance and the emergence of multidrug-resistant parasite. So I investigated the genetic diversity of Korean vivax malaria using 4 different types of antigenic genes: ((merozoite surface proteins (PvMSP), circumsporozoite proteins (PvCSP), apical membrane antigens (PvAMA), and Duffy binding protein (PvDBP). These markers were highly polymorphic and have been used as markers in P. vivax population studies. In addition, genotyping protocol based on analyzing different 4 types of genes would be helpful to distinguish between new infection and treatment failure cases or domestic infection and imported cases. A total of 156 blood samples from recent vivax patients in Korea were genotyped by PCR reaction and sequenced. As results, six MSP-1 subtypes (S-a, S-b, S-c, B-1, B-2, Re), four different AMA-1 subtypes (N1, N2, N3, N4), five subtypes (haplotypes) of CSP VK210 and four DBP subtypes (Type-1, Type-2, Type-3, Typ-4). Among these, N2 of AMA-1 and Type-2 subtypes of DBP were newly found in this study. Analysis of all the data combined by genotyping methods of 4 different genes revealed 60 different combination sets genotypes among 156 analyzed samples. Then ‘S-b, subK-2, N2, type-4’ combination is predominant sets (16%) in South Korean currently. On the other hand, 33 patients were unique as each one of them was found in a single isolate only and it means that there are many genotypes of parasite quite variety in South Korea. But, these combination sets did not presented remarkable characteristics related to geographical diversities or time span. In summary, the polymorphism of the 4 different antigenic genes (MSP-1, CSP, AMA-1 and DBP) could be considered suitable genetic markers for the analysis of P. vivax population in South Korea. The protocol associate molecular techniques for P. vivax genotyping, which is low cost method based on highly polymorphic gene, a highly discriminatory genetic tool, which provides complementary genotyping data, if required. In addition to genotyping protocol based on analyzing different 4 types of genes would be applied to distinguish between new infection and treatment failure or domestic infection and imported cases. The methodology can be easily used as moderate- or high- throughput analysis protocol for clinical trials or epidemiological studies.
