Pentopan, Promozyme, Celluclast, Ultraflo, Pectinex, Ceremix, Viscozyme 및 Tunicase 등의 다양한 탄수화물분해효소 중 인삼 잎, 줄기 및 뿌리에 처리하였을때 고형분 수율과 총당 수율이 가장 높은 Ceremix를 선별하였다. 실험계획법을 이용하여 효소처리 조건을 최적화한 결과 인삼 잎, 줄기 및 뿌리를 0.15 ㎜의 크기로 분쇄하여 사용하고, pH 5.0-5.5와 55-60℃에서 Cremix를 1%의 농도로 첨가하여 2시간 동안 효소처리하였을 때 높은 총당 추출 수율을 얻을 수 있었다. 인삼 잎, 줄기 및 뿌리의 효소처리군의 TLC 분석 결과, 효소처리한 인삼 뿌리 실험군은 인삼의 다당류가 올리고당류까지 분해된 것을 알 수 있었다. 또한 효소처리한 인삼 잎, 줄기 및 뿌리의 총올리고당 함량은 효소처리하지 않은 인삼 잎, 줄기 및 뿌리의 추출물에 비해 총 올리고당 함량이 각각 2.2배, 5.3배 및 2.3배 증가하였다. 인삼의 잎, 줄기 및 뿌리의 효소처리군이 효소비처리군에 비해 유산균 증식이 활성화되어 총올리고당의 소모속도와 산도의 변화가 큰 것으로 나타났다. 인삼의 잎, 줄기 및 뿌리를 무처리, 효소처리에 비해 발효처리, 효소처리 후 발효처리하였을 경우 인삼사포닌의 대사산물 중 각각 Compound K, Rg1, Rh1 및 Rg3의 특정사포닌이 선택적으로 증가하였으며, 그 중 잎의 경우에는 C-K가 무처리군에 비해 효소처리군에서 3배정도 증가하였고, 발효처리, 효소처리 후 발효처리군에서는 10배 이상 선택적으로 증가하는 것을 알 수 있었다.

The extraction and enzymatic treatment conditions of ginseng leaves, stems and roots for the production of fermented ginseng were optimized in order to enhance the extraction of oligosaccharide, which is a lactobacillus growth-activating factor. Additionally, the effects of enzymatic hydrolysis on lactobacillus growth and ginsenoside metabolite formation were investigated. The ginseng leaves were found to be more suitable for the raw material of fermented ginseng products because ginseng leaves have higher carbohydrate and crude saponin contents than ginseng roots. Among eight carbohydrate hydrolysis enzymes including Pentopan, Promozyme, Celluclast, Ultraflo, Pectinex, Ceremix, Viscozyme and Tunicase, which were tested for the hydrolysis of ginseng leaves, stems and roots, Ceremix gave the highest dry matter and total saccharide yields. The enzymatic hydrolysis conditions were statistically optimized by an experimental design method. The optimized conditions were found as particle size of ginseng raw material below 0.15 ㎜, pH 5.0-5.5, reaction temperature of 55-60℃, Ceremix concentration of 1%, and reaction time of 2 hr. It was shown that the polysaccharides of ginseng were hydrolysed to oligosaccharide by the enzymatic hydrolysis of ginseng leaves, stems and roots. The total oligosaccharide contents increased by the enzyme treatment up to 2.2-fold, 5.3-fold and 2.3-fold, respectively compared to control (no treatment). It was found that the enzymatic treatment promoted the lactobacillus growth, resulting in more significant change in total oligosaccharide consumption and total acidity. The contents of several ginsenoside metabolite compounds such as Compound K, Rg1, Rh1 and Rg3 were selectively increased by combining the enzymatic treatment and lactobacillus fermentation. Especially, in case of enzyme treatment using ginseng leaves, Compound K formation was enhanced up to 3-fold compared to control (no treatment). Moreover, in case of combined treatment of enzyme and fermentation, Compound K formation was significantly promoted up to 10-fold.
