The aim of the present work is to compare and characterize chemical composition, morphology, and surface roughness of cp-Ti surfaces modified by grit-blasting and acid etching and to observe the relation with cell behavior. Hydroxyapatite (HA) and aluminum oxide (Al2O3) granules were used as the blasting material. And HNO3 and H3PO4 were used as the etching agent. The morphology of Ti implant surface was characterized using SEM, the surface roughness was measured by laser scanning surface profilometer, and the surface chemistry were examined using EDS and XRD. Cell viability and proliferation tests were conducted and compared by MTT assay, in terms of the blasting and etching conditions. The surface roughness test shows that the samples blasted with larger grit size obtained higher average surface roughness (Ra), and the samples blasted with Al2O3 have higher roughness (8.555 ?m, grit size of 100-150 ?m) than that with blasted with HA (7.291 ?m). The acid etching process reduced the surface roughness of the HA blasted samples (from 7.291 ?m to 5.806 ?m and 5.627 ?m by H3PO4 and HNO3, respectively), and reduced slightly the Al2O3 blasted samples (from 8.555 ?m to 8.267 ?m and 7.943 ?m by H3PO4 and HNO3, respectively). The EDS result showed the presence of Ca and P atoms on the HA blasted Ti disc surface. Both samples blasted with HA and Al2O3 grits showed the significant amount of grit residues. After the H3PO4 and HNO3 etched samples, the HA residues were almost removed from the surface. However, the Al2O3 residues were almost remained on the Ti surface. These results were confirmed with XRD studies. Overall, all samples (polished, blasted and blasted/etched) showed the good cell viability and cell proliferation. The cell proliferation measured by MTT assay increased with the period of cell culture; after 2 hours (40.75 OD), 1 day (66.18 OD) and 1 week (79.10 OD) on HA blasted samples, and after 2 hours (32.57 OD), 1 day (42.53 OD) and 1 week (47.67 OD) on Al2O3 blasted samples. After 1 week, the smoother surface samples blasted with smaller grit size (blasted and blasted/etched samples) revealed slightly higher cell proliferation than the rougher surface samples. However, the cell proliferation of Al2O3 blasted samples was lower than that of HA blasted samples due to the residual Al2O3 grits embedded on the surface.
