본 연구에서는 항균력이 우수한 에센셜오일을 피부 관리에 적용 시 항균 효과가 있는 에센셜오일이 함유하고 있는 자극 성분에 의해 피부 부작용을 일으킬 수 있는 가능성이 있으므로, 좀 더 안전하게 부작용이 적으면서 미용 분야에 사용할 수 있는 오일들의 필요성을 인식하여 연구를 진행하게 되었다. 자극적인 성분이 적으면서 항균, 항진균 효과가 우수하다고 보고된 에센셜오일 중에서 기존의 활용도가 높은 오일 외에 사용 빈도가 낮은 편인 제라늄과 팔마로사를 중심으로 본 연구를 하게 되었다. 본 연구에서는 제라늄과 팔마로사, 두 에센셜오일의 주성분인 geraniol, geranyl acetate, β-citronellol를 구입하여 항균 실험과 인체에 적용하기 위한 연구를 할 때 행해지는 항산화 실험을 진행하였다. 에센셜오일은 천연물이므로 원산지나 허브의 수확 시기에 따라 성분 함량이 일정치 않으므로 HPLC를 이용하여 실험에 사용되는 제라늄과 팔마로사 성분 분석을 하고, 각 성분의 함량에 따른 제라늄과 팔마로사의 항산화, 항균력과의 연관성을 알아보았다. 제라늄과 팔마로사를 분석하기 전 geraniol, geranyl acetate, β-citronellol을 분석하여 이 성분들이 검출되는 시간과 제라늄과 팔마로사를 분석했을 때 나오는 geraniol, geranyl acetate, β-citronellol이 검출되는 시간을 비교하여 서로 동일한 물질임을 확인하고, 두 에센셜오일의 각 성분 함량을 측정하였다. 실험 결과 제라늄은 geraniol을 46.27%, β-citronellol을 30.44%, geranyl acetate를 1.68% 함유하였고, 팔마로사는 geraniol을 72.55%, geranyl acetate를 25.26% 함유하였다. 항산화 효과를 알아보기 위한 DPPH radical 소거 효과를 이용한 항산화 실험에서 geraniol, geranyl acetate, β-citronellol는 항산화 효과를 보이지 않았고, 팔마로사는 낮은 항산화 효과를 보였고, 제라늄은 10% 농도에서 대조군인 L-ascorbic acid 보다 높은 항산화 효과를 보였다. Geraniol, geranyl acetate, β-citronellol의 단일 시료보다 여러 화학 성분이 복합되어진 제라늄과 팔마로사의 항산화 효과가 더 우수한 것은 어느 한 성분의 영향을 받기보다 여러 복합 성분의 영향으로 항산화 효과가 있을 것으로 여겨진다. 항균력을 보기 위한 Paper disc 기법을 이용한 실험에서 geraniol과 팔마로사는 Candida albicans, Propionibacterium acnes, Staphylococcus aureus, Escherichia coli에 항균 효과를 나타내었다. 실험에 사용된 각 시료 농도가 1%일 때 geraniol이 팔마로사 보다 항균력이 높았고, 각 시료 농도가 10%일 때 팔마로사가 geraniol 보다 항균력이 높게 나타났다. 1% 에서는 geraniol의 함량이 팔마로사의 항균력에 영향을 미치지만, 10%일 경우에는 geraniol의 영향보다는 다른 미량 성분들과의 복합적인 활성으로 팔마로사가 geraniol 보다 높은 항균력을 나타내었다. In this study, the investigation started from necessity on essential oils which have both antimicrobial activity and useful effects in beauty industry with less side effects. Essential oils with excellent antimicrobial activity may cause skin irritations in skin care due to the irritant ingredients which these essential oils have. Among essential oils reported with less irritation and better antimicrobial and antifungal activities, this study chose Pelagonium graveolens (Geranium) and Cymbopogon martini (Palmarosa) which are less used than other well-known and frequently used essential oils for further investigation. The study investigated antioxidative activity test like as studies of application to body in cosmetics or skin care and antimicrobial activity test by purchasing the two essential oils-Geranium and Palmarosa, and the main ingredients of these-geraniol, geranyl acetate, β-citronellol. In this study, it was assumed that essential oils may have differences in composition of ingredients according to their origins because oils are natural products. For this reason, analysis on composition by HPLC was fulfilled and also study to find out relation between antioxidative activities and antimicrobial activities of Geranium and Palmarosa by contents of ingredients was investigated. This study showed that the main ingredients of Geranium and Palmarosa - geraniol, geranyl acetate, β-citronellol are the same substances by comparing detection times of each ingredients. Also the composition of these ingredients in these two essential oils was detected. As a result, Geranium had 46.27% of geraniol, 30.44% of β-citronellol and 1.68% of geranyl acetate, and Palmarosa had 72.55% of geraniol and 25.26% of geranyl acetate. And DPPH radical scavenging activity for antioxidative activity test showed no antioxidative activity in each of geraniol, geranyl acetate, and β-citronellol, low antioxidative activity in Geranium, and higher antioxidative activity in 10% solution of Palmarosa comparing to control group of L-ascorbic acid. The antioxidative activity in complex of these main ingredients in Geranium or Palmarosa was much better than the activity of single ingredients, and it meant that the antioxidative activity might come not from single ingredients' activity but from complex of these ingrendients. In the antimicrobial activity test by Paper disc method, geraniol and Palmarosa showed antimicrobial activities on Candida albicans, Propionibacterium acnes, Staphylococcus aureus, Escherichia coli. Antimicrobial activity of geraniol was higher than that of Palmarosa in 1% solution, but when it comes to 10% solution, Palmarosa showed better antimicrobial acitivity than geraniol. And it menat that composition of geraniol affected antimicrobial activity of Palmarosa in 1% solution, but in 10% solution, compositive activation of other small quantity of ingredients with geraniol affected the better antimicrobial activity in Palmarosa. Geranyl acetate showed antimicrobial activity only to Escherichia coli. In 1% solution of β-citronellol showed antimicrobial activity on Propionibacterium acnes, and in 10% solution of Geranium and β-citronellol showed the activity on Candida albicans, Propionibacterium acnes, Staphylococcus aureus. Geranium in antioxidative activity test, and Palmarosa in antimicrobial activity test showed excellent effects, and this result meant that it was more effective to use Geranium or Palmarosa than to use individual ingredients. Palmarosa showed antimicrobial activity in 1% solution against Propionibacterium acnes, and Geranium showed activity in 10% solution. This meant that both essential oils could be useful for Acne skin care by antimicrobial activity on Propionibacterium acnes. This study in antimicrobial activity of Geranium and Palmarosa against certain bacteria such as Candida albicans, Propionibacterium acnes, Staphylococcus aureus, Escherichia coli which may cause skin trouble can help to use of Geranium and Palmarosa in troubled skin care for improvement of skin condition.
