HERG channels 에서 ginsenoside Rg3은 농도, 전압에 의존적으로 steady-state IHERG, peak tail (Itail), 그리고 deactivating tail currents (Iend-tail) 모두 활성되는 것으로 나타났다. 이 각각에 EC50 값은 0.41 ± 0.05, 0.61 ± 0.11 그리고 0.36 ± 0.04 µM 으로 나타났다. In the current study, we sought to identify these site(s) in Xenopus oocytes expressing wild-type and mutant L(α1C)-type Ca2+ channels using the two-microelectrode voltage-clamp technique. To this end, we first assessed how various point mutations of the L-type Ca2+ channel affected the action of Rg3. Mutations L427R, N428R and L431K in transmembrane domain-1-segment 6 (IS6) of the channel reduced the effect of Rg3. Rg3 treatment produced a negative shift in the inactivation voltage but did not alter the steady-state activation voltage, and none of the mutant channels affected the Rg3-induced negative shift of inactivation voltage (Section I). In the present study, we examined the effects of ginsenosides on rat brain BKCa (rSlo) channel heterologously expressed in Xenopus oocytes to elucidate the molecular mechanisms how ginsenosides modulate the excitability of neuronal cells. Ginsenoside Rg3 (Rg3) activated outward BKCa channel activity. Rg3-mediated enhancement of outward BKCa channel currents was concentration-dependent, voltage-dependent and reversible manners. Rg3 action was independent of intracellular Ca2+. The EC50 was 15.1 ± 3.1 M. The mutation Y360I abolished the Rg3 action as well as Rg3-mediated rightward shift of TEA concentration-response curve (Section II).
