본 연구에서는 인간 keratinocyte cell line, HaCaT에서 분화 상태에 따른 beta-defensin 3의 상이한 발현 조절과 이에 대한 cyclooxygenase-2 (COX-2) 의 연관성을 밝히고자 하였다. beta-defensin 3는 인간의 피부에서 발현되는 중요한 천연의 항균 펩타이드 (anti-microbial peptide)의 일종으로 아토피 피부염 (atopic dermatitis)에서 만성적인 피부질환의 악화와 병원성 미생물의 비정상적인 아토피 피부 병변의 천이에 병리적 원인이 되며, 이는 피부를 구성하는 각질형성세포 (keratinocyte)가 발현하는 항균 펩타이드 이다. 본 연구에서는 다양한 피부 생물학 및 아토피 피부염 같은 피부 병리현상을 연구하는데 있어 가장 많이 사용하는 인간 각질형성세포주인 HaCaT에서 Ca2+-의존성 분화 및 탈분화 기전을 이용하여 각질형성세포의 분화 상태에 따른 beta-defensin 3의 발현 상태 및 조절 기전을 연구하였다. HaCaT은 배양 배지 내 칼슘의 제거와 첨가로 가역적으로 탈분화 및 분화 상태의 유도가 가능하였으며, 이를 활용하여 분화 상태의 HaCaT에서 beta-defensin 3의 발현이 크게 증가함을 확인하였다. 또한 분화와 탈분화된 HaCaT의 cDNA micro array 분석 연구를 통하여 다양한 차이 유전자들 중 염증성 단백질인 COX-2 유전자의 발현이 분화 상태에 따른 HaCaT에서 beta-defensin 3의 조절에 중요한 매개 단백질임을 real-time PCR과 immunoblotting 실험을 통해 확인하였다. 이러한 결과들을 바탕으로 다양한 외래 미생물에 대한 1차 방어막으로 작용하는 천연의 항균 펩타이드 beta-defensin 3의 발현이 외부 유래의 다양한 미생물 혹은 항원 물질과 접촉의 가능성이 많은 피부 최외각에서 그 발현양이 증가하며, 이는 또한 분화된 피부각질형성세포에서 염증 반응성의 일환인 증대 된 COX-2의 발현과 연관되어 있음을 확인하였다.

I have studied to identify a regulatory role of differentiation stage on the expression of human beta-defensin 3 and involement of cyclooxygenase-2 (COX-2) in keratinocyte cell line, HaCaT. Human beta-defensin 3 is expressed in human skin and is regarded as key anti-microbial peptide. Insufficient expression of beta-defensin 3 from epidermal keratinocytes often results in abnormal colonization of pathological microorganisms in atopic dermatitis. Using HaCaT cell line which was most widely used in studies physiology and pathology such as atopic dermatitis of skin and Ca2+ -dependent differentiation and de-differentiation process, I investigated a regulatory mechanism of differentiation on the expression of beta-defensin 3. By adding or depleting Ca2+ in culture medium, HaCaT was reversibly differentiated or de-differentiated. When HaCaT was in the differentiated stage, the expression of beta-defensin 3 was dramatically increased when compared with de-differentiated HaCaT. To further study any possible cellular mechanisms controlling the identified elevated expression of beta-defensin 3 only in differentiated HaCaT, I tried cDNA microarray analysis to compare genomic profiles between de-differentiated HaCaT and differentiated HaCaT. Among significantly changed genes, COX-2 but not COX-1 was identified and its involvement to the identified regulation of beta-defensin 3 was confirmed by various experiments. Based on the identified results, I assumed that beta-defensin 3 working as primary defense mechanism against invading microorganisms was elevated in terminally differentiated HaCaT where have much more chances to meet the mircoorganisms than in de-differentiated HaCaT. Also, the elevation of beta-defensin 3 was tightly regulated by inflamatory-related protein, COX-2 in differentiated HaCaT.
