Phytochemical constituents were isolated from the CHCl3 fraction of Hallabong ([Citrus unshiu Marcov. × C. sinensis Osbeck] × C. reticulata Blanco) peels through open column chromatography and MeOH recrystallization. And, we simultaneous determined to part-specific preponderance of polymethoxyflavones by HPLC analysis. Moreover, we evaluated potential of naturally occurring aldose reductase (AR) inhibitors in the peels of hallabong using rat lens AR. First, the sliced and dried peels of hallabong extracted with MeOH under reflux and concentrated in vacuo to give MeOH extracts. The extracts thus obtained were suspended in distilled water and then partitioned in turn using n-Hx, CHCl3, EtOAc, and n-BuOH. A portion of the CHCl3 fraction was chromatographed on a silica gel column using a stepwise gradient of n-Hx-EtOAc and EtOAc-MeOH solvent system to yield sub-fractions. Through recrystallization, compounds were yielded. Their structures were identified 5,6,7,3′,4′-pentamethoxyflavone (1), 6,7,8,3′,4′-pentamethoxyflavone (2), 3-hydroxy-5,6,7,4′-tetramethoxy flavones (3), 5,6,7,8,3′,4′-hexamethoxyflavone (4), 3,6,7,4′-tetramethoxyflavone (5), 3,5,6,7,8,3′,4′-heptamethoxyflavone (6), and 5,6,7,8,4′-pentamethoxyflavone (7) based on comparison of the spectral data (1H-, 13C-NMR and MS) with data from the literature. Among these compounds, 3,6,7,4′-tetramethoxyflavone (5) was isolated for the first time from nature. Second, the simultaneously determination for the part-specific preponderance of PMFs was performed with the peels, leaves and stems of hallabong, as by-products, by HPLC analysis. Standards were isolated and identified from previous experiment. The C18 column and mixture of water and ACN were used for the simultaneous determination of PMFs. The gradient elution program was 15 to 55% ACN within 45 min. The UV spectra were recorded 330 nm for the identification. The major PMFs of hallabong are 3,6,7,4′-tetramethoxyflavone (5) in the peels (15.4 mg/g) and 5,6,7,8,4′-pentamethoxyflavone (7) in the leaves (12.2 mg/g). Through total contents of PMFs in by-products from hallabong, the content of the leaves and peels (30.0 and 26.5 mg/g) are more high than that of the stems. Third, we evaluated potential of naturally occurring AR inhibitors in the peels of hallabong using rat lens AR. Above all, the assay for AR inhibition activity was tested in extract level with MeOH extract and four fractions of hallabong. The CHCl3 fraction has good AR inhibitory activity (84.65%). Next, this assay was tested in compounds level with seven flavonoids. Compounds 2, 4, 5, and 7 were demonstrated AR inhibitory activity.
