Plasticity and self-renewal capacity of BMCs offer huge potential for clinical tissue regeneration. Recently, Bone marrow derived stem cells can transdifferentiate into multilineage cells, such as muscle of mesoderm, lung and liver of endoderm, pigment cell of ectoderm origin, and germ cell. However, mechanism and evidence of differentiation of BMCs into germ cells is still unclear and no study attempt to generate BMCs derived progenies. Several types of cells such as blastoderm cell, primodial germ cells and embryonic germ cells were injected into early stage of recipient embryos for producing chimera avian and understanding cell development purpose. However, no results about avian BMCs related with developmental study are reported yet. Therefore, this study is to assess chicken BMCs (chBMC) differentiation ability into multi lineged cells and capability to generate chimera and donor BMCs derived progenies. BMCs were isolated from eGFP (enhanced green fluorescent protein) transgenic and Barred Plymouth Rock (BPR) roosters. Then, BMCs were separated by percoll density gradient (90, 45, and 0%) method. Among 3 layers of cell population, expressions of stem cell specific transcript gene such as Nanog, Sox2, Pou V, mesenchymal stem cell specific markers (THY1, CD44), and positive reaction for AP staining were observed in middle layered cells by RT-PCR and AP staining analysis. The FACS revealed that 17 % of cells were positively reacted with the stem cell surface-specific marker antibody, SSEA-4. When isolated chBMCs were cultured for 30 days with retinoic acid supplements, they were reacted with 3 germ-layered-specific antibodies (ectoderm: Tuj1, mesoderm: brachury, endoderm: α-fetoprotein) and male germ cell specific markers (Vasa, c-kit and Dazl). Isolated BMCs from eGFP Tg rooster were transferred into testes of busulfan treated recipients. Localization and in vivo trans-differentiation of injected cells in the seminiferous tubules of recipients were traced for up to 40 days post-injection by GFP expression and immunocytochemical analyses. The integration of the eGFP and the neoR genes in sperm gDNAs of recipient was confirmed via PCR analysis. Transferred BMCs were survived and proliferated more than 40 days in seminiferous tubules of recipients. Immunocytochemistry analysis indicated that transferred BMCs were differentiated into germ cell in testes of recipients. The sperm quality parameters such as concentration, volume, viability, total sperm numbers, and fertilization rate of ejaculated spermatozoa from recipients were assessed and the semen of the BMC-transferred recipients was inferior in all aspects of the parameters. Ratio of eGFP positive spermatozoa in ejaculated semen of recipient was between 0.01 and 0.82%. A subsequent testcross of the recipient roosters with non-transgenic hens resulted in the production of eGFP transgenic progenies, demonstrating the successful trans-differentiation of the adult BMC to the germ cells in the testis. In addition, approximately 5,000 ~ 6,000 BPR BMCs were injected into the blastoderm of white leghorn (WL) recipients. Developmental and hatching rates of BMCs injected eggs were significantly lower than those of normal eggs. Somatic chimera chicks were generated with 2.7% frequency (2 of 73) using BMCs injection methods. Confirmation of chimerism in hatched chicks was achieved with PCR analysis using D-loop specific primers of BPR and WL. Therefore, this study demonstrated the successful production of the chimera chicks using chBMC. Taken together, these results suggest that the use of adult BMCs is a new and promising approach to the production of transgenic poultry, and may prove helpful in the study of avian developmental biology.
