It was also determined the effect of apple-pomace extracts on laccase activity and its mRNA expression in the mycelium of P. ostreatus. Enzyme activity and its mRNA level were greatly increased 5 fold and 2 fold with the addition of apple pomace extracts, respectively. To examine changes in the metabolic reaction of mycelium to the treatment of apple-pomace extracts, extracellular and intracellular proteins of mycelium in liquid culture medium were analyzed by using SDS-PAGE and 2-D gel electrophoresis. As the result of SDS-PAGE analysis, several bands in the sample of extracellular proteins were observed and their band intensities were dependent on the treatment concentration of apple-pomace extracts. Analysis of 2-D gel electrophoresis with the intracellular proteins of mycelium showed that 300 protein spots were observed and 7 spots were greatly changed in their densities. One of seven spots was increased and the others were decreased in density levels by the treatment of apple-pomace extracts. In conclusion, apple-pomace extracts has greatly induced laccase enzyme activity and laccase gene expression in the mycelium of mushroom.
