본 연구는 돌외로부터 주요 gypenoside를 다량 순수 분리하고, 다양한 처리 조건
과 효소 처리을 통해 고부가가치의 인삼 사포닌으로 전환 최적 조건과 전환 기술을
개발하는데 그 목적이 있다. 돌외 사포닌 중 인삼 사포닌으로 전환 가능한
protopanxadiol 계열 사포닌을 탐색하였고, 그 중 돌외에 다량 함유되어 있는

켰을 때 최종 산물로 compound K를 생산하는 것을 LC/TOF/MS 분석으로 확인
하였고, HPLC를 이용하여 gypenoside V가 ginsenoside Rd를 거쳐 F2로 전환되
고 최종적으로 compound K를 생성하는 전환 경로임을 알 수 있었다. Viscozyme
L과 gypenoside V를 반응시켰을 때 최종산물로 ginsenoside F2를 생산하는 것을
LC/TOF/MS로 확인 하였고, ginsenoside Rd를 거쳐 F2로 전환되는 경로를 HPLC
로 정량적으로 확인하였다. Viscozyme L을 이용 하였을 경우 Citrozym ultra L보
다 gypenoside V를 더 빠른 시간, 더 높은 수율로서 ginsenoside F2를 생성하는
것으로 보아 효소 활성이 더 높은 것으로 사료 되었다. 반면 gypenoside V에
Citrozym ultra L를 처리 한 경우, 반응 최종 생산물로 compound K를 생성하는
전환 경로를 나타내었다. 이러한 효소학적 특징들을 이용하여 인삼 이외의 다른 식
물인 돌외로부터 고기능성, 고부가 가치의 목적하는 ginsenoside를 선택적으로 대
량 생산 할 수 있다면 경제적으로 더 유용한 자원활용의 예가 될 것으로 사료된다.

In the process of hydrolyzing rhamnopyranoside and glucopyranoside from gypenoside V by various transformation methods such as heating, acid, enzyme treatment, and the gypenoside V was converted to the minor ginsenoside which has more pharmaceutical effects.
Gypenoside V was transformed to ginsenoside Rg3 and Rh2 in the mild acid condition. The transformation of ginsenoside Rg3 and Rh2 was reacted by increasing reaction temperature and time in the acidic condition, the optimal reaction time and temperature for this bioconversion was 3hrs and 80℃. Consequently, we confirmed transformation of the gypenoside V to minor ginsenosides by TLC and HPLC analysis. The inherent bioconversion pathway was evaluated as follows, gypenoside V → ginsenoside Rg3 → Rh2.
Gypenoside V was also bioconverted into the ginsenoside F2 and compound K by food grade enzymes, Citrozym ultra L and Viscozyme L, respectively. These enzymes have α-rhamnosidase and β-glucosidase activity, converted gypenoside V to ginsenoside Rd, F2 and compound K. The resultant metabolites and their molecular weight were identified by LC/TOF/MS. The enzymatic reaction was analyzed by HPLC, suggesting the conversion pathway as follows gypenoside V → ginsenoside Rd → F2 → compound K.
We proposed the optimum condition for the gypenoside V conversion by heating, acid, and enzyme treatment. These process would allow the specific bioconversion of gypenoside V to various ginsenosides using an approprate heating, acid, and enzyme treatment. These results that suggest that it is possible to make highly functional minor ginsenosides from Gynostemma pentaphyllum.
