The iPSCs make up for the weak points somatic stem cells and embryonic stem cells, and provides an opportunity to generate patient- or disease-specific pluripotent stem cells. However, most iPS cells are made by viral vectors, such as retrovirus (Okita et al., 2007; Takahashi and Yamanaka, 2006; Varas et al., 2009; Wernig et al., 2007) and lentivirus (Takahashi et al., 2007; Winkler et al., 2010), which integrate the exogenous reprogramming factors into host genomes and may increase the risk of tumor formation. Previous studies demonstrated that transient expression of Cre recombinase try to delete reprogramming factors (Kaji et al., 2009; Soldner et al., 2009). However, they have still incomplement deletion and hazard of secondary integration. Here, we used a Cre-loxP system in order to remove the exogenous reprogramming factors. First we produce a Cre protein to prevent the integration of nucleic acid after a reprogramming. After iPSC was treated with Cre protein, we identified to elimination of the transgenes that induced the stemless of fibroblast cells and observed mOrange-negative iPSC colonies through laser confocal microscope. iPSCs deleted exogenous reprogramming factors have clearly characteristic of iPSCs. Those results suggested that Cre recombinant proteins show the potential beneficial effects in the elimination of integrated reprogramming factors, compare to viral infection.
