Mitogen-activated protein kinase (MAPK) pathway is major signaling that cellular molecular network maintains delicate balance, which is regulated during inflammatory responses. Several groups of evidence demonstrate that phosphorylation of MAPKs in the signal transduction pathway plays critical roles on downstream events of inflammatory response. The MAPK pathway is regulated by protein phosphorylation and dephosphorylation. Protein tyrosine phosphatases (PTPs) play critical roles in MAPK signaling by dephospho- rylation of the MAPK subfamily, including the c-Jun N-terminal kinase (JNK), extracellular signal-regulated kinase (ERK), and p38. This study suggests that PTPs mediate TNF-α secretion through regulation of MAPK activity. Preferentially sixty-five PTP expressions were examined in mouse macrophage, RAW264.7 cells by RT-PCR. Based on this result, the relationship between PTP expression and TNF-α production was verified in LPS stimulated RAW264.7 cells by TNF-α ELISA. Consequently, PTPε, PTPH1, and DUSP26 reduce LPS-induced TNF-α production in RAW264.7 cells. In addition, PTP expression levels by LPS stimuli were determined RT-PCR. mRNA expression level of PTPH1 was induced by LPS but PTPε and DUSP26 were not. In addition, we tested whether PTPε, PTPH1, and DUSP26 regulate activation of MAPK family. RAW264.7 cells that were transfected with PTPε, PTPH1, and DUSP26 genes after LPS treatment were analyzed by immunoblotting. Consequently, PTPε reduces LPS-induced p-38 activation while DUSP26 inhibit p38 and JNK activation by LPS. PTPH1, however, has no effect on MAPKs. Taken together, this study suggests that PTPε and DUSP26 inhibit LPS induced TNF-α production through inactivation of MAPKs in RAW264.7 cells. PTPH1, also, reduces the TNF-α secretion by LPS and might be involved in other cellular signaling pathway. Therefore, this study could be experimental basis for identification of novel targets as anti-inflammatory agents in various immune diseases.
