At first, I studied whether PTPs are involved in the regulation of pro-inflammatory cytokines in RAW 264.7 mouse macrophage cells stimulated with lipopolysaccharide (LPS). The mRNA and protein expression levels of PTPN7 and DUSP5 were significantly regulated under LPS stimulation. PTPN7 and DUSP5 expression suppressed the production of pro-inflammatory cytokines in LPS-stimulated RAW 264.7 cells. Besides, up-regulation of PTPN7 and DUSP5 lead to inhibition of signal transduction pathway. PTPN7 has a negative regulatory function to extracellular signal regulated kinase 1/2 (ERK1/2) and p38 and DUSP5 inhibits ERK1/2 and NF-κB signals. At second, I showed how DUSP6 regulates p53-dependent apoptosis. This study demonstrates that DUSP6 binds to and activates p53 and then induces p21 expression, which is a p53 target gene. In addition, DUSP6 certainly inhibited phosphorylation levels of Ser-376 in p53, which has known as p53 inactivation sites. Thus, DUSP6 could achieve induction cleavage of PARP and caspase-3, known as apoptosis-associated proteins, by modulating p53 dephosphorylation. Therefore, this study could be experimental basis for identification of novel targets as anti-inflammatory agents in various immune diseases.
