Chitin in crustacean cuticles is tightly associated with inorganic salts such as calcium carbonate, proteins, and lipids, including pigments. To isolate chitin from crustacean shells, three steps are required, namely, demineralization (DM), deproteinization (DP), and elimination of lipids, including pigments. The traditional method for chitin production was performed through a chemical process using inorganic acids for DM and strong alkali for DP. As an alternative to the chemical process, biological production of chitin using organic acid-producing bacteria (Latobacillsu paracasei KCTC-3074, Lactobacillus plantarum KCTC-13093) and protease-producing bacteria (Serratia marcescens FS-3) could reduce a source of environmental pollution and the depolymerization of chitin. The protein content analysis method by the nitrogen content method of measurement was very useful as the method of measurement of the protein which in crab shells. In KCTC 3074+FS-3 (1:1) cofermentation, the level of demineralization was the highest value of 97.2%, but the level of deproteinization in the cofermentation was 52.6% at day 7. Protein content in the treatment of FS-3 alone reduced from 22.4 to 3.62%. The successive fermentation in the combination of FS-3 and KCTC-3074 gave the best result in co-removal of CaCO_(3) and proteins from crab shell. In this combination, the rates of demineralization and deproteinization were 94.3% and 68.9%, respectively, at the end of fermentation. Compared with that of without pretreatment of crab shell, Following a pretreatment by a combined HCl and Sonication method, the crab shell were separately treated by KCTC-13093 and Serratia marcescens FS-3. It was found that the pretreatment of crab shell could help the DM increased from 91% to 95% by KCTC-13093 and could help the DP increased from 77% to 87% by FS-3. As to treatment by biological treatment, the recovery of chitin and chitosan was higher than the chemical treatment, were 20.1 g, 17.5 g, respectively.
