작물의 생물학적 방제에 사용되는 생물농약소재를 발굴하기 위하여 국내의 곤충병원성 선충으로부터 살충성 공생세균을 확보하고 이로부터 살충성 유전자원의 구성과 특징을 확인하기 위한 연구를 수행하였다. 본 논문에서 얻은 결과는 다음과 같다. 1. 공생세균의 분리 및 동정 국내의 살충성 공생세균과 이들의 유전자원 확보를 위하여 토양 유래의 선충으로부터 꿀벌부채명나방 유충에 살충력을 나타내는 곤충병원성 선충을 확보하였다. 이들로부터 공생세균의 특성을 이용하여 MacConkey와 NBTA 배지를 이용하여 공생세균을 선택적으로 분리하였다. 먼저, MacConkey agar 배지에서 그람 음성 간균을 분리한 다음, 그중에서 bromothymol blue를 흡착하고 tetrazolium chloride를 환원하는 공생세균을 NBTA agar 배지로부터 분리하였다. 분리한 공생세균의 동정은 16S rDNA 염기서열을 이용하였다. 그 결과 NBTA 배지에서 짙은 녹색 또는 짙은 노란색을 띠는 5 균주의 공생세균은 모두 Photorhabdus temperata로 확인되었다. 이들은 type strain인 P. temperata subsp. temperata XINach strain과 99% 상동성을 나타내었고, 16S rDNA 서열에서 Photorhabdus와 Xenorhabdus 속을 구분지는 서열인 ‘TGAAAG' 및 제한효소 EcoR I site를 확인하였다. 2. 분리한 공생세균의 생장 및 생리학적 특성 5종의 P. temperata의 생장과 생리학적 특성을 조사한 결과, 그람 음성 간균이며 catalase 양성반응을 나타내었다. 그리고 TSB 배지에서 짙은 주황색의 색소를 생성하며 skim milk와 Tween-20 또는 80을 각각 함유한 배지에서 protease 및 lipase 활성을 나타내었다. 또한, Salmonella typhimurium과 Micrococcus luteus에 대한 항균 활성을 확인하였다. 지방산 조성 및 함량도 기존에 보고된 P. temperata와 유사한 pattern을 나타내었다. 이들의 생장 특성을 조사한 결과 배양 5∼6일째에 최대 균체 생장을 나타내었으며, 대부분 대수 증식기 기간이 매우 긴 것으로 나타났다. 또한, 균주 생장 시 필요한 yeast extract의 적정 농도는 3∼5%로 나타났으며 생장 최적 온도는 28∼30℃로 확인되었다. 5종의 P. temperata의 생장제한온도는 35∼ 36℃로 나타났다. 3. 공생세균의 살충력 및 배양상징액의 내열성 검증 분리한 5종의 P. temperata의 살충력을 확인하기 위하여 이미 살충력이 검증된 P. luminescens TT01 균주를 표준 균주로 사용하였으며, 배양시간별 배양상징액을 5령 이상의 꿀벌부채명나방 유충에 주사하였다. 그 결과 5종의 P. temperata와 1종의 표준 균주 모두 유충에 대한 높은 살충력을 나타내었다. 살충력은 대수 증식기 후반의 배양상징액이 가장 높았으며, 특히 P. temperata M1021과 J5 균주는 주사 시 마비증상과 함께 주사 후 1일 만에 100% 살충력을 나타내었다. 가장 높은 살충력을 나타낸 96시간 배양상징액의 내열성을 알아보기 위하여 40∼100℃의 온도에서 30분간 열처리한 다음 유충에 주사하여 살충력을 확인하였다. J5와 M1021 균주는 80℃에서 30분간 열처리한 후에도 살충력이 대부분 유지되었으며, 90℃에서도 50% 이상의 살충력을 나타내었다. 그러나 TT01 균주를 포함한 4 균주는 열처리 온도가 증가할수록 살충력이 급격하게 감소하였다. 이러한 내열성 살충력을 나타내는 살충성분의 단백질 유무를 확인하기 위하여 배양상징액에 proteinase K를 처리하여 단백질 활성을 제거하였다. 이를 다시 꿀벌부채명나방 유충에 주사한 결과, J6 균주와 TT01 균주는 살충력이 각각 60%와 50%를 나타내었으며 나머지 4 균주는 살충력의 80% 이상이 유지되었다. Proteinase K에 의해 단백질 대부분이 제거되었음에도 높은 온도에서 살충력을 가지는 것으로 보아 비단백질성 내열성 살충 성분의 존재를 확인할 수 있었다. 4. 동결건조법을 이용한 Photorhabdus temperata M1021의 제제화 내열성 살충력이 가장 우수한 P. temperata M1021 균주를 동결건조법을 이용하여 제제화한 다음 저장 안정성 및 살충력 실험을 하여 이들의 생물농약 소재로서의 제품화 가능성을 조사하였다. 동결건조 시 세포의 손실을 최소화하기 위하여 skim milk, starch, sodium alginate, dextrose와 sodium glutamate를 보호제로 첨가하였다. 동결건조 시료에서의 균의 생존율은 skim milk를 7%(w/v) 첨가하였을 때 63%로 가장 높았으며 그 이상의 농도에서는 감소하였다. 그러나 다른 보호제는 동결건조 시 세포보호능력이 매우 낮은 것으로 확인되었다. 가장 높은 생존율을 나타낸 skim milk 첨가 동결건조 시료는, skim milk 7%일 때 저장 안정성이 75% 이상(4주)으로 나타났으나 저장기간이 증가함에 따라 점차 감소하였다. 그러나 동결건조분말에 대한 주사 독성은 2.02 × 101 세포 이상에서는 주사 후 4일 이내에 100% 살충력을 나타내었으며, 2.02 × 100 세포와 같이 아주 적은 농도에서도 유충의 살충효과가 50%를 나타내었다. 5. 독소 복합체 확보를 위한 genomic cosmid library 제작 및 분석 살충성 공생세균인 P. temperata M1021로부터 이들의 살충 활성에 관여하는 독소 복합체 및 독소 단백질의 존재와 구성을 확인하고 이들의 살충력을 확인하고자 먼저, P. temperata M1021 균주의 genomic cosmid library(PtC)를 구축하여 이들의 유전자원을 확보하였다. 구축한 library로부터 pooling-subpooling PCR 법을 이용하여 독소 유전자를 포함하는 4개의 cosmid clone을 확보하였다. 확보한 cosmid clone의 plasmid 염기서열을 분석한 결과, 독소 복합체인 tcd locus와 tcc locus에 해당하는 7종의 독소 유전자를 확보하였다. 상동성 분석 결과 기존에 보고된 P. luminescens 및 P. asymbiotica의 독소 복합체 살충 단백질과는 많은 차이를 나타내었다. 6. 독소 복합체의 살충력 검증을 위한 재조합 발현시스템 구축 및 발현 상기에서 확보한 독소 복합체의 살충력을 검증하고자 T7 promoter를 가지는 pET21a(+) 발현 벡터를 이용하여 재조합 발현시스템을 구축하였다. 그 결과 TcdA-like를 제외한 대부분의 재조합 단백질은 IPTG에 의해서만 발현되었다. 그러나 TcaC와 TccC-like를 제외한 대부분의 재조합 단백질은 불용성 형태로 전환되었다. 이러한 재조합 단백질의 용해도를 증가시키기 위하여 TccB, TcdB2와 TccC-like를 대상으로 온도, IPTG 농도 및 배지 종류를 변화시켜 목적 단백질의 발현형태를 확인하였다. 그러나 발현 강도에만 영향을 줄 뿐 단백질의 용해도 증가에는 영향을 주지 않았다. 또한, 불용성 형태의 refolding 과정에서도 용해도는 증가하지 않았다. 마지막으로 arabinose promoter를 가진 pEAra 발현벡터를 제작하여 목적 단백질의 용해도를 증가시키고자 하였다. 각각의 독소 component인 TccB, TcdB2와 TccC-like의 발현 유무를 확인한 결과, TccB는 15℃와 27℃에서 일부 soluble form으로 발현되었으며, TcdB2는 15℃에서 대부분 soluble form으로 발현되었다. 목적 단백질의 정제에는 Ni-Sepharose FF resin을 사용하여 정제하였으나 TcdB2만 정제되었다. 7. 재조합 독소 복합체의 살충력 검증 pET 발현 시스템에 의해 목적 단백질이 발현된 재조합 대장균의 살충력은 꿀벌부채명나방과 갈색거저리 유충에 대한 주사 독성으로 확인하였다. 꿀벌부채명나방 유충의 경우 TccB가 발현된 재조합 대장균이 가장 높은 살충력을 나타내었으며 주사 후 5일 때에 유충의 살충력은 85%를 나타내었다. TccC-like와 TccC는 각각 37.5%와 52.5%의 살충력을 나타내었다. 갈색거저리 유충의 경우 TccB가 발현된 대장균을 주사한 경우 가장 높은 살충력을 나타내었고, 주사 후 5일 때에 100% 살충력을 나타내었다. 그리고 TccC-like는 57.5%, TcdB2가 35%의 살충력을 나타내었다. pEAra 발현 시스템은 TccB와 TcdB2의 꿀벌부채명나방 유충에 대한 살충력 검증에는 목적 단백질이 발현된 대장균 세포를 파쇄하여 유충에 주입하였다. 그 결과 TccB가 발현된 대장균을 주사한 후 2일 만에 유충이 모두 죽었지만 TcdB2가 발현된 대장균과 정제된 TcdB2는 유충에 대한 살충 효과가 나타나지 않았다. 8. 독소 복합체 및 살충성 유전자원 확보를 위한 염기서열 분석 마지막으로 P. temperata M1021 균주의 cosmid library로부터 확보한 독소 복합체 관련 유전자 이외에, 나머지 독소 복합체 locus와 그 외의 독소 유전자 및 항균·항생물질 관련 유전 정보를 더욱 더 효율적으로 확보하기 위하여 차세대 염기서열시험법(NGS)을 이용하여 염기서열 분석을 하였다. 이들로부터 확보한 357개의 contig를 re-assembly 하여 최종적으로 237개의 contig를 확보하였으며 이들에 대한 annotation 작업을 수행하였다. 그 결과 tcb locus를 제외한 tca, tcc, tcd locus의 존재와 구성을 확인하였으며, 그밖에 mcf locus와 여러 개의 cytotoxin, 항균 및 항생물질 합성에 관여하는 수많은 NRPS의 존재도 확인하였다. 현재, 그 유전 정보가 알려지지 않은 P. temperata에 대한 유전정보를 분석하고 해독하여 관련 정보를 제공함에 따라 기존에 보고된 Photorhabdus 속과의 genome 비교가 가능할 것으로 여겨진다.

Entomopathogenic bacteria such as Photorhabdus spp. are virulent pathogen of a wide range of insect larvae when entomopathogenic nematodes invade the larvae of susceptible insects, Photorhabdus spp., living in the gut of the nematodes, are released into the insect hemolymph and produce a variety of toxins that are causes of death of the victim. In order to develop the Photorhabdus as biological control agent for agriculture, entomopathogenic nematodes were collected from soil of local forest and several entomopathogenic symbiotic bacteria were isolated. Five Photorhabdus temperata strains showing thermostable toxicity against Galleria mellonella larvae were isolated. To develop the strains for biological agents, P. temperata M1021, showed one of the best toxicity from the isolates, was lyophilized. The freeze-dried bacterial cell was tested its viability, storage stability and insecticidal ability. The genomic cosmid library of the strain M1021 was constructed genomic DNA sequencing was performed to obtain the genetic resources of toxin complexes, insecticidal toxins and antibiotics. In this paper, the results obtained are as follows: 1. Isolation and identification of symbiotic bacteria from entomopathogenic nematodes Five symbiotic bacteria were isolated from the entomopathogenic nematode, Heterorhabditis sp., which were collected from soil samples in various fields in Korea. All isolates produced bright-pink to red colonies when they were on MacConkey agar media. Upon NBTA agar, primary colonies showed mucoid, convex and greenish or dark yellow colonies with a clear zone around the colonies due to bromothymol blue absorption and tetrazolium chloride reduction. The 16S rDNA of the five isolates were sequenced in a length of 1,505 nucleotides. Phylogenetic analysis of 16S rDNA showed that bacteria can be classified as Photorhabdus temperata. Sequence alignment with those of Photorhabdus spp. indicated that the five isolates had the highest sequence similarity with Photorhabdus temperata subsp. temperata. Five of symbiotic bacteria J3, J5, J6, J7 and M1021 were identified as P. temperata. A characteristic sequence of Photorhabdus was found in 199-204 as ‘TGAAAG’. 2. Growth and physiological characteristics of isolated symbiotic bacteria The growth conditions of the isolates like culture temperature, media, physiological characteristics as phenotype, pigmentation, catalase, lipase, proteinase and antibiotic production were investigated. All of the isolates had catalase activity and they were shown to be gram negative. They showed orange pigments in TSB media, lipase activity on Tween 20∼80, and protease activity on skim milk. In addition, they exhibited antimicrobial activity against Salmonella typhimurium and Micrococcus luteus. The growth of the five bacterial isolates and reference strains P. luminescens TT01 showed the long exponential phase of 2∼4 days and stationary phase of 5∼6 days. The growth conditions were optimized at 3∼5% of yeast extract and 28∼30℃ of temperature, they didn’t grow above 35℃ or 36℃. 3. Verification of the insecticidal activity and heat stability of culture supernatant The culture supernatant of isolates showed high insecticidal activity against Galleria mellonella, especially when the culture was harvested at late exponential growth phase (3∼4 days). The heat stability of insecticidal activity was tested by heat treatments for 30 min at various temperature range. The supernatants extracted from the cultures of P. temperata J5 and M1021 showed 95% of remaining insecticidal activity after heat treatment at 80℃ for 30 min. The insecticidal activities of all culture supernatants were remained after proteinase K treatment. 4. Insecticidal activity of freeze-dried P. temperata M1021 P. temperata M1021 was lyophilized using the freeze-drying. Protective agents such as skim milk, starch, sodium alginate, glucose and sodium alginate of various concentrations were tested to protect cells during freeze-drying and confirmed the cell viability after freeze-drying. Lyophilized P. temperata M1021 containing skim milk 7% (w/v) showed the highest survival rate of 63%. While other protective agents had no effect on the viability of the cells. The freeze-dried powder made of 7% (w/v) skim milk containing P. temperata M1021 showed good survival stability (above 75%) for 4 weeks at 4℃. Exhibited marked insecticidal activity was marked when the lyophilized cell were injected to larvae of G. mellonella. All of the G. mellonella larvae were killed within 2 days post infection at a dose of above 2.02 × 101 cells of P. temperata M1021. Construction and analysis of genomic cosmid library to screen the toxin complexes A genomic cosmid library was constructed to assure the existence and composition of toxin genes. Cosmid clones including toxin genes obtained from the library by using pooling-subpooling PCR method. Purified cosmids from the clones were sequenced, analysed and verified. Seven species of toxin gene, applicable to toxin complex tcd locus and tcc locus were found. 6. Production and validation of recombinant toxin proteins To validate the insecticidal activity of toxin protein, a the recombinant expression system using pET21a(+) was constructed. The result showed that the most of recombinant proteins were expressed by IPTG except from TcdA-like. But the most part of recombinant protein was found from insoluble fraction except TcaC and TccC-like. To increase the solubility of the recombinant toxin protein, various expression conditions were tried under different temperature, IPTG concentration, and media. But the solubility of the recombinant protein was not improved in any cases. When the promoter of the expression vector was changed from T7 promoter to E. coli araBAD promoter, the solubilities of the recombinant protein TccB and TcdB2 were increased at 15℃ or 27℃. 7. Bioassay of recombinant toxin complex protein The toxicity of E. coli harboring toxin genes was tested by hemocoel injection into larvae of G. mellonella and Tenebrio molitor. E. coli expressing recombinant TccB showed high toxicity after 5 days of injection to G. mellonella larvae. Eighty five percent of larvae were died by TccB expressing E. coli. In case of TccC-like and TccC protein, 37.5% and 52.5% of mortality to G. mellonella larvae were shown, respectively. All of the T. molitor larvae were died after 5 days of injection of E. coli expressing TccB. Mortalities of TccC-like and TcdB2 protein toward T. molitor larvae were showed 57.5% and 35%, respectively. When concentrated crude extract of E. coli harboring TccB gene was injected to T. molitor larvae, all of the victim were died after only two days. While extract of E. coli harboring TcdB2 protein didn't show the insecticidal activity against larvae of G. mellonella. 8. Genome sequencing of P. temperata M1021 for securing genetic resources of toxin complexes and insecticidal components In order to genetic resources of toxin complexes, insecticidal components and antibacterial components, full genomic sequence analysis was performed. Total 237 contigs were obtained after annotation of the raw sequence data. From the contigs, tca, tcc, and tcd locus as well as mcf locus, multiple cytotoxin and numerous NRPS (Non-ribosomal peptide synthases) involved in the synthesis of antibacterial and antibiotic, were found. Genetic information related the insecticidal activity and pathogenicity of P. temperata M1021 will be useful for development of biological control agent.
Entomopathogenic bacteria such as Photorhabdus spp. are virulent pathogen of a wide range of insect larvae when entomopathogenic nematodes invade the larvae of susceptible insects, Photorhabdus spp., living in the gut of the nematodes, are released into the insect hemolymph and produce a variety of toxins that are causes of death of the victim. In order to develop the Photorhabdus as biological control agent for agriculture, entomopathogenic nematodes were collected from soil of local forest and several entomopathogenic symbiotic bacteria were isolated. Five Photorhabdus temperata strains showing thermostable toxicity against Galleria mellonella larvae were isolated. To develop the strains for biological agents, P. temperata M1021, showed one of the best toxicity from the isolates, was lyophilized. The freeze-dried bacterial cell was tested its viability, storage stability and insecticidal ability. The genomic cosmid library of the strain M1021 was constructed genomic DNA sequencing was performed to obtain the genetic resources of toxin complexes, insecticidal toxins and antibiotics. In this paper, the results obtained are as follows: 1. Isolation and identification of symbiotic bacteria from entomopathogenic nematodes Five symbiotic bacteria were isolated from the entomopathogenic nematode, Heterorhabditis sp., which were collected from soil samples in various fields in Korea. All isolates produced bright-pink to red colonies when they were on MacConkey agar media. Upon NBTA agar, primary colonies showed mucoid, convex and greenish or dark yellow colonies with a clear zone around the colonies due to bromothymol blue absorption and tetrazolium chloride reduction. The 16S rDNA of the five isolates were sequenced in a length of 1,505 nucleotides. Phylogenetic analysis of 16S rDNA showed that bacteria can be classified as Photorhabdus temperata. Sequence alignment with those of Photorhabdus spp. indicated that the five isolates had the highest sequence similarity with Photorhabdus temperata subsp. temperata. Five of symbiotic bacteria J3, J5, J6, J7 and M1021 were identified as P. temperata. A characteristic sequence of Photorhabdus was found in 199-204 as ‘TGAAAG’. 2. Growth and physiological characteristics of isolated symbiotic bacteria The growth conditions of the isolates like culture temperature, media, physiological characteristics as phenotype, pigmentation, catalase, lipase, proteinase and antibiotic production were investigated. All of the isolates had catalase activity and they were shown to be gram negative. They showed orange pigments in TSB media, lipase activity on Tween 20∼80, and protease activity on skim milk. In addition, they exhibited antimicrobial activity against Salmonella typhimurium and Micrococcus luteus.
