The second category is abiotic stresses, and it includes heat, low temperatures (chilling, freezing), UV, salinity, ozone, pesticides, hypoxia, air pollution and heavy metal. Among these, specially low temperature and salinity are known to affect severely in plant growth and crop yields. Plants respond to abiotic stresses through a range of different strategies, including physiological and biochemical processes that are normally mediated by the up-regulation of the expression of stress-regulated genes. Previously we tried to elucidate the signal transduction pathways by which plants resist cold stress (<0℃) using hot pepper (Capsicum annuum L.) as a model plant, and then we demonstrated how this knowledge can be used to develop cold resistance crop plants. We were able to select four cold stress related transcription factors : a family of ethylene responsive element binding protein (CaEREBP-C1, -C2, -C3) and wizz-like transcription factor (CaWRKY1). Here, we constructed plant overexpression vectors of these four genes into pB7WG2D using the Gateway system, and transformed tobacco plants by Agrobacterium-mediated transformation method. Insertions of each gene were confirmed by genomic PCR. About 30 individual transformant lines for each gene were evaluated for expression level of the introduced genes by Northern blot analysis. The resultant transgenic tobocco plants which harbors cold-stress related gene for transcription factor show strong tolerance to cold stress.
