고로쇠 추출물의 화장품, 식품, 의약외품 및 의약품 등 여러 분야에서의 활용 가능성을 알아보기 위해 항산화 효과를 연구하였다. 추출물은 고로쇠 가지의 목피와 목질부를 3가지 용매(ethanol, ethyl acetate, distilled water)를 사용하여 얻었으며, 수액은 천연 상태를 사용하였다. 고로쇠 추출물의 수율은 목피 추출물이 7.5∼14.4%, 목질부 추출물이 3.3∼5.5 %로 나타났으며, 수액은 2.5%의 수율을 나타냈다. 고로쇠 추출물 중에서 water로 추출한 목피 14.4%, 목질부 5.5%로 높게 나타났다. 고로쇠 추출물을 분석한 결과, 고로쇠 추출물의 총 폴리페놀 함량은 1.5∼47.7 mg/g이었고, 총 플라보노이드 함량은 0.08∼13.1 mg/g 이었다. 고로쇠 추출물의 폴리페놀 성분은 phenolic acid계인 tannic acid, 3,5- dihydroxybenzoic acid가 주성분인 것으로 확인되었다. 고로쇠 추출물의 초기 반응에 있어서 DPPH free radical 소거 속도와 대조군 BHT인 IC50 기준에서의 DPPH 소거 속도를 분석함으로써, 고로쇠 추출물 중에서 목피와 목질부 추출물들이 대조군 BHT보다 상대적으로 빠른 DPPH 소거능을 나타냈으며, 수액은 미미한 DPPH 소거능을 나타냈다. 고로쇠 추출물들의 반응 속도 기준으로 DPPH free radical 소거능을 분석함으로써, DPPH 소거 속도가 우수한 목피와 목질부 추출물이 항산화 효과가 있음을 확인하였다.

Acer mono max. extracts were obtained from bark and xylem of branch part using three kind of solvent(ethanol, ethyl acetate, distilled water), and sap is used as natural state. The yields were shown that bark extracts were ranged from 7.5 to 14.4% and xylem extracts were ranged from 3.3 to 5.5 %. The yield of sap was 2.5%. Distilled water extracts of bark had a higher yield as 14.4% than any other extracts among Acer mono max. extracts. In the component analysis of Acer mono Max. extracts, total polyphenol content were ranged from 1.5 to 47.7 mg/g and total flavonoid content were ranged from 0.08 to 13.1 mg/g. Tannic acid and 3,5-dihydroxybenzoic acid of phenolic compound were identified as main component in Acer mono max. extracts. Antioxidative activities were determined using DPPH as a free radical. In the results of determination on condition that DPPH free radical scavenging rate in initial reaction rate and DPPH scavenging rate by BHT IC50 value, bark extracts and xylem extracts indicated relatively stronger DPPH scavenging activities than BHT and sap. DPPH scvenging rate of sap was indicated insignificant activity. In addition, DPPH scavenging activities were increased concentration- dependently. It showed positive correlation between antioxidative activities and kinetics reaction rate(kinetic mechanism). According to the results obtained in the present study, it suggests that bark extracts and xylem extracts of Acer mono Max. have potential antioxidative activity and can be considered as an effective antioxidant.
