다음 제브라피쉬 embryo에서 당뇨모델을 수립하기 위한 validation 실험을 수행하였다. 여러 농도 (5, 10, 25, 100 uM)의 2-NBDG 용액에 노출시킨 뒤 (0.5, 1, 2 h) alloxan 용액 (100, 300, 500, 700 uM)으로 옮겨 노출시킨다 (0.25, 0.5, 1, 2 h). 실험결과, 2-NBDG로 전처리한 제브라피쉬 embryo에서 alloxan의 영향으로 pancreatic islet의 크기가 감소하였다. 25 uM의 2-NBDG에 1 시간 동안 노출시킨 뒤 500 uM 의 alloxan에 30 분 동안 처리하여 제브라피쉬 embryo 당뇨모델을 수립하였다. 수립된 당뇨모델이 당뇨모델로서 이용 가능한가를 확인하기 위하여 홍삼과 ginsenoside Rg1, Rg3, Rb1을 처리하여 효능을 확인하였다. 본 연구를 통하여 수립된 모델들이 앞으로 당뇨모델로 사용될 수 있을것이라고 사료된다.

In this study, at first, it was performed a validation study for the establishment of optimal hyperglycemia involving the following factors: alloxan concentrations of 100, 200, 300 and 400 mg/100 ml, exposure times of 30, 60, 120 and 180 minutes to an alloxan solution and glucose solution concentrations of 1%, 2%, and 3%. The results have shown that exposure to glucose solution following alloxan induction might increase blood glucose level in zebrafish in a dose- and time-dependent manner.
