다양한 생활성 물질의 표면 위에서 특정 작용기들이 직접 맞닿아 상호작용이 일어난다. 이를테면 효소와 기질의 결합, 약물과 표적과의 기작 등, 모두 표면에 존재 하는 작용기에 의존한다. 특히 표면에서 원하는 위치에 원하는 물질의 반응을 유도함으로써 다양한 생체 내 물질의 유무를 확인하는 센서 혹은 약물을 효율적으로 운반할 수 있는 운반체의 개발에 응용될 수 있다. 또한, 생체유사표면을 재현함으로써 다양한 생명 현상 연구의 수행이 가능하다. 따라서, 생활성 표면의 분석 및 생활성 물질의 표면 정량은 생명공학분야에서 그 중요성이 최근 들어 더욱 조명되어 오고 있는 주제로서 많은 연구가 진행되어 오고 있다. 이 논문에서는 표면에 존재하는 단백질의 정성과 정량분석에 대한 두 가지 연구 주제에 대해 소개하겠다. 첫 번째 주제는 금 표면을 이용한 자기조립단분자층(SAMs)과 매트릭스 보조 레이저 탈착 비행시간형 질량분석기를 이용하여 앱타머와 표적 단백질간의 결합 친화력을 확인하는 연구이다. 표적 단백질로 급성 중증 호흡기 증후군 헬리케이즈를 사용하였다. 금 표면에 10% 헬리케이즈 앱타머를 고정시키고 90%는 oligo(ethylene glycol)로 고정 시켰다. 헬리케이즈 앱타머가 고정된 표면에 표적단백질인 헬리케이즈를 처리하여 표면에 존재하는 표적 단백질을 트립신(단백질분해효소)에 의해 분해하고 그 결과물인 펩타이드를 매트릭스 보조 레이저 탈착 비행시간형 질량분석기로 분석하였다. 이전에 알려진 앱타머와 표적 단백질 간의 결합 친화력을 확인하는 전기영동적 이동성 분석, 나이트로셀룰로스 필터 결합 분석 등과 같은 방법들은 시간이 많이 소요되고 시료의 양이 많아야 한다는 단점을 가지고 있으며, 이 연구는 이를 극복 할 수 있는 방안으로 제시 될 수 있다. 두 번째 주제는 금 나노 입자 표면위에 존재하는 단백질을 정량하는 연구이다. 금 나노입자 표면에 단백질을 코팅 혹은 고정을 시킨 후 입자 표면에 존재하는 단백질을 트립신에 의해 분해하고 그 결과물인 펩타이드는 동위원소가 있는 내위표준물질을 가한 후 매트릭스 보조 레이저 탈착 비행시간형 질량분석기로 정량했다. 내위표준물질은 분해된 표적 단백질의 대표펩타이드와 서열이 같고 다른 분자량을 가진다. 금 나노 입자 표면에 존재하는 단백질의 절대적인 양은 대표펩타이드와 내위표준물질과의 질량분석기 신호 세기를 비교함으로써 정량할 수 있다. 이 두 분석물의 질량 신호 세기는 같은 분자 환경을 갖고 있기 때문에 금 나노 입자에서의 분석물의 실제 양을 나타낸다. 현재까지는 2차원표면에서의 생체분자을 확인하는 방법으로 효소 면역측정법, 진동 수정 마이크로저울, 표면 플라즈몬 공명과 같은 방법들이 존재하지만 3차원 입자의 표면에서 효과적인 절대 정량방법은 보고된 바가 드물다. 우리가 개발한 3차원 표면에서의 절대 정량법은 질병치료, 약물전달시스템 등과 같은 연구들을 수행하는 데 있어서 표면 생체분자의 정량적 정보를 제공함으로써 연구의 성패를 결정하는 중요한 요인으로 작용될 것이다.

Analysis and quantification of surface-bound biomolecules are important in the field of biotechnology. Therefore, analytical tools for various surface’ characterizations have been actively studied and developed. In this study, I will introduce two parts of my study about identification and quantification of surface-bound proteins using proteolysis and matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS). First, I report on a method for validation of different affinity between RNA aptamers and a target protein using self assembled monolayer (SAMs) and MALDI-TOF MS. RNA aptamers against Severe Acute Respiratory Syndrome (SARS) coronavirus helicase (SCV helicase) were employed as a target protein. Our approach uses gold substrate which possesses the RNA aptamer with 10% surface density amongst 90% of oligo (ethylene glycol) moieties. Helicase is captured on the RNA aptamer-presenting monolayers, subsequently digested by trypsin and analyzed by MALDI-TOF MS. Aptamer-presenting monolayers and MALDI-TOF MS analysis clearly discriminated stronger binding against weaker binding to the target protein. In the second part, I describe quantification of proteins on three dimensional surfaces. Protein-coated nanoparticles have been used in many studies such as drug delivery, disease diagnosis, therapeutics, and bioassay. In those applications, amount and density of proteins on a particles’ surface are important parameters that need to be calculable. While quantification methods for two dimensional surface-bound proteins are common such as surface plasmon resonance (SPR), quartz crystal microbalance (QCM) measurements, and enzyme-linked immunosorbent assay (ELISA), efficient methods for quantification of proteins on three dimensional surfaces such as nanoparticles have not been reported yet. In this study, the immobilized proteins on the AuNPs are digested by trypsin, and the resulting peptide fragments are analyzed by MALDI-TOF MS in the presence of an isotope-labeled internal standard (IS). IS has the same sequence as reference peptide of bovine serum albumin (BSA) digestion but different molecular weight. Comparing the mass intensities between a reference peptide from the protein and IS enables absolute quantification of the surface-bound protein. The intensity of these two analytes represents the real amounts of analytes on the AuNPs, because they have the same molecular milieu.
