This study was aimed to evaluate organic removal and methane production using food wastes as a substrate in a two-phase anaerobic digestion system of alternating mesophilic acidogenic-mesophilic methanogenic (system A), mesophilic acidogenic-thermophilic methanogenic (system B), and thermophilic acidogenic- mesophilic methanogenic (system C). Furthermore, changes in the microbial community structure within the reactors were identified using 16s rDNA gene amplification and T-RFLP analysis. By changing the operating temperature of acidogenic to methanogenic reactor, more than 3 HRT's were required to stabilize the system due to inhibition of methanogenic bacteria from the low pH feed from the acidogenic reactor. The highest TCOD removal of 86.4% was recorded in system B while system C has the highest SCOD removal of 96.6%. Comparing system A to system B, the TS removal was increased by 0.5%, while in system C a decrease of 0.1 % TS removal was observed relative to system A. This could suggest that the difference of TS removal was not high in each system. However, VS removal in system A, B, and C, were 78.5%, 81.7%, and 79.2%, respectively. System B did not only exhibit high VS removal but also have the highest methane gas production of 0.44 L CH4/gVSadded and methane content of 70.7%(v/v). Changes in the structure of microbial communities were identified according to the changes in the operating conditions of the reactor through the T-RFLP analysis.
