Use of this fungus in practical biocontrol requires the production of large amounts of conidia. The B. bassiana KK5 strain, which has high virulence to green peach aphid (Myzus persicae), was studied to produce conidia by solid-state fermentation (SSF). The optimal condition for conidial production on the polished white rice was found to be 40% moisture content of rice, 25C of culture temperature, 2-day-old seeding culture grown in a liquid medium composed of 3% corn meal, 2% rice bran, and 2% corn steep powder, 107conidia g-1 of initial conidia concentration in the inoculated rice, 10% inoculum size, polyethylene bag as a container of rice during cultivation. Using the optimal condition, the maximum conidial production, 4.05 g conidia/100 g dry rice was achieved at the 14th day of cultivation and the yield was 2.83 times higher than that of pre-optimization. For extending the shelf life of conidia of B. bassiana, the conidia were dried to moisture content less than 5%. The germination ratio of the dried conidia stored at three different temperatures (4, 25, and 35C) during 9 months storage showed that the conidia stored at 4C exhibited the highest viability (85.9%). Conidial viability tests were conducted with different combinations of oil, emulsifier and thickener. Hereupon, 16 oils, 6 emulsifiers, and 4 thickeners were used for formulation of conidia. Some oils (SSTH, SST3H, MO 840, Isopar M, diesel and mineral heavy oil), emulsifiers (Tween 80, PLE 3, EFW), thickeners (AT 50, HFS) showing relatively higer conidia viability were chosen for the oil-based formulation of conidia. And then, the effects of different combinations of the oil, emulsifier and thickener on the viability of conidia during 9 months storage at 25C were evaluated. The conidial viability in all formulations tended to be slowly declined over the storage time. After storage for 9 months at 25C, the viability of conidia in M8 and IM oil-based formulations containing EFW as emulsifier and HFS as thickener were the highest with 42.5 3.2% and 44.4 5.6%, respectively, among the tested formulations. The combined formulation or application of a fungal entomopathogen and a plant extract with insecticidal activity may improve aphid control efficacy and reduce application concentration. Therefore, the effect of plant extracts such as melia (Melia azedarach var. japonica), neem oil and six different commercial plant extracts were evaluated for their influence on the germination of conidia in oil-based formulation. The results showed that all the eight plant extracts reduced or retarded conidia germination, so these plant extracts could not be mixed with conidia in oil-based formulation for long-term storage. However, these plant extracts could be mixed with conidia in oil-based formulation in application suspension just before immediate spraying. Combined effect of conidia in oil-based formulation and plant extract was evaluated against M. persicae by detached leaf test.
