1. 녹용 부위별 추출물의 수율은 100℃에서 추출하였을 가장 높았고, 잔사를 이용하여 추출한 추출물에서도 일부 유효성분이 추출되었다. 반면 70% 에탄올을 이용하여 추출한 결과 열수 추출물보다는 수율이 낮았다.
2. 생리활성 성분인 uronic acid, sulfated-GAGs 및 sialic acid의 함량을 측정한 결과, uronic acid의 경우는 100℃ 추출물에서 가장 높은 함량을 나타내었고 잔사를 이용한 추출물에서도 일부 함량이 나타났다. 또한 부위별로는 상대에서 가장 높았고 하대로 내려갈수록 함량이 낮았다. 반면 70% 에탄올 추출물의 경우는 열수 추출물보다 월등히 높은 uronic acid 함량을 나타내어 용매에 따른 유효성분 추출이 다름을 알 수 있었다. Sulfated-GAGs의 경우는 열수 추출물의 함량이 월등히 우수하였고 70% 에탄올 추출물에서는 아주 소량만 검출되었다. 부위별로는 상대 추출물에서 함량이 높았고 중대 및 하대에서는 그 함량이 낮았다. 온도가 높아질수록 sulfated-GAGs 함량이 줄어들었지만 잔사를 이용한 것을 감안하면 함량이 높음을 알수 있었다. 따라서 sulfated-GAGs의 추출은 온도에 영향을 많이 받음을 알 수 있었다. Sialic acid의 경우는 70% 에탄올 추출물에서는 검출되지 않았으며 열수 추출물의 경우는 110℃ 추출물의 함량이 가장 높아 역시 온도에 영향을 많이 받음을 알 수 있었고, 부위별로는 상대와 중대 추출물의 전체 함량은 비슷하였고 하대는 조금 낮았다.
3. 추출물의 항산화 효과는 DPPH, hydroxyl, hydrogen peroxide 소거 활성 및 환원력을 측정하여 평가하였다. 추출물의 DPPH 소거 활성은 열수 및 70% 에탄올 추출물 모두에서 소거 활성이 나타났고 각 부위별 100℃ 추출물에서 가장 높은 소거 활성을 나타내었다. 그러나 부위별로 비교한 결과 DPPH 소거 활성은 별다른 차이가 없었다. Hydrogen peroxide 소거 활성은 열수 추출물의 경우 온도가 높을수록 소거 활성이 약간 증가하는 경향을 나타내었고 70% 에탄올 추출물이 열수 추출물보다 우수한 소거 활성을 나타내었다. 그러나 부위별 소거 활성은 거의 유사하였다. Hydroxyl 라디칼 소거 활성은 70% 에탄올 추출물이 아주 우수한 소거 활성을 나타내었다. 반면 열수 추출물은 100℃ 추출물에서 가장 높은 소거 활성을 나타내었고 온도가 올라갈수록 활성은 감소하였다. 역시 부위별 소거 활성은 모두 비슷하게 나타났다. 녹용 추출물들이 환원력은 상대 추출물에서 가장 우수하였고 하대로 갈수록 활성은 감소하였다. 또한 온도가 상승할수록 활성이 감소하였고, 각 부위별로 70% 에탄올 추출물의 환원력이 열수 추출물보다 우수하였다.

(Abstract) The velvet antler cut from deers, which belong to the Cervidae, is generally termed Nokyong in Korea and has been long recognized as one of the most effective and powerful invigorants, as well as an enhancer of systemic resistance to infectious diseases. It is widely used as a traditional oriental medicine in Korea and China, with their consumption being associated with anti-stress, anti-aging, anti-inflammatory effects. In this study, velvet antler was divided into upper, middle and base sections, and was subjected to the extraction process using boiling water at three different temperature (100℃, 110℃ and 120℃) and 70% EtOH solution. Among the extracts, the yield was highest in the extracts with each sections on 100℃ and the yield was decreased with increasing temperature. Biologically active substances such as uronic acid, sulfated-glycosaminoglycans (GAGs) and sialic acid in the extracts were analyzed, and antioxidant activities of the extracts were investigated. Uronic acid contents in hot water extracts significantly decreased with increasing temperature (p < 0.05) and the extract obtained from the upper section possessed significantly higher uronic acid content than those of the extracts obtained from the middle and the base sections. In other side, uronic acid contents of the 70% EtOH extracts in each section were significantly higher than those of the extracts by hot water. And also uronic acid content was highest in the upper section, followed by the middle and base sections. Sulfated-GAGs contents of each extract by hot water decreased with increasing temperature and the extract obtained from the upper section showed higher level of sulfated-GAGs than those of the extracts obtained from the middle and the base sections. No significant difference observed between the extract of the middle and base sections. Furthermore, 70% EtOH extracts in each section showed lower sulfated-GAGs contents compared to the hot water extracts. Sialic acid contents of each extract by hot water showed different pattern compared to the uronic acid and sulfated-GAGs. Overall sialic acid content was highest in the upper section and the extracts at 110°C showed highest content. However, no sialic acid was observed in the extracts with 70% EtOH. Antioxidant activity of velvet antler extracts was evaluated using scavenging ability toward DPPH, hydroxyl radical, hydrogen peroxide, and reducing power. Velvet antler extracts obtained from each section effectively quenched DPPH radical in a dose-dependent. The DPPH scavenging activity was decreased with increasing temperature and the 100°C extracts exhibited the highest scavenging activity in each section. The 70% EtOH extracts in each section also showed DPPH radical scavenging activity in a dose-dependent, and the activity was lower than that of hot water extracts at 100°C. No significant difference (p < 0.05) between each section extracts was observed. Hydrogen peroxide scavenging activity was augmented with increasing concentrations. All hot water extracts of velvet antler displayed similar activities while the 70% EtOH extracts showed comparatively higher activity.
