In the present study, using an experimental model of LPS-induced endotoxin shock, the protective effects and mechanisms of AJ were investigated. Mice were administrated intraperitoneally with AJ (0.1, 0.5 and 1 g/kg) and then after 1 h, intraperitoneally injected with LPS (37.5 mg/kg) of lethal dose. The survival rate was examined per every 12 h. For further study of organ damage, 3 h after LPS injection, the mice were sacrificed and then the lung and liver tissue were harvested. The administration of AJ inhibited LPS-induced endotoxin shock. AJ treatment inhibited lung damage and inflammation such as neutrophil infiltration, and production of interleukin (IL)-1b, IL-6, tumor necrosis factor (TNF)-a, and type-I interferon (IFN). AJ treatment also inhibited the liver damage, neutrophil infiltration, and the production of type-I IFN, but not of IL-1b, IL-6, and TNF-a in liver. In peritoneal macrophages, AJ also inhibited LPS-induced production of type-I IFN. In addition, AJ reduced the activation of signal transducer and activator of transcription(STAT)-1 protein signal. In conclusion, these results suggest that AJ inhibits LPS-induced endotoxin shock and organ damage by inhibiting the production of type-I IFN. AJ might be a beneficial effect on inflammatory diseases such as sepsis.
