Cudrania tricuspidata, an herbal plant widely used in East Asia, possesses antioxidant, antibacterial, and anti-inflammatory activities. To identify the active compounds in C. tricuspidata, prenylated xanthones have been isolated. These xanthone derivatives were found to have α-glucosidase inhibitory activity. I therefore determined whether extracts of C. tricuspidata classified according to its parts have distinct effects in α-glucosidase inhibitory assay. For this in vitro assay, I isolated the roots, leaves, and stems of the plants before extraction the plants were also divided into groups according to harvest time. I determined inhibitor type of samples by creating a Lineweaver-Burk plot. Consequently stem extracts were more effective inhibitor on α-glucosidase than leaf extracts. Root extract from plants harvested in January, which had 77% velocity inhibition on 300 μg/mL and 41.6 μg/mL of inhibitory constant, was the most effective inhibitor. Inhibitory percentage of positive control at 1 mM was approximately 67% of enzymatic velocity with acarbose and 41% velocity with rosiglitazone. According to the Michaelis-Menten equation, the type mechanism underlying the effects of stem and root samples was competitive or noncompetitive inhibition. Additionally, acarbose was found to be a mixed-type noncompetitive inhibitor, and rosiglitazone was classified as a competitive inhibitor. These results suggest that C. tricuspidata, a widely used food resource, can serve as an anti-diabetic biomaterial.
