Flaniostatin의 항산화 활성, 세포독성을 확인하고 tyrosinase 저해 활성을 측정함으로서 Flaniostatin이 새로운 피부미백제가 될 가능성을 제시하였다.

Tyrosinase is known to be the first, two and rate-limiting enzyme in the synthesis of melanin pigments responsible for colouring skin. Tyrosinase inhibition is one major strategy used to treat hyperpigmentation. In human skin melanocytes, the cellular tyrosinase inhibition was examined by the conversion of L-tyrosine and oxidation of L-DOPA to DOPAquinone. We evaluated the tyrosinase inhibitory effects with in vitro systems to find skin-whitening agents without cytotoxic concerns. We have found that the methanol extract of dried skin of Cudrania tricuspidata shows potent tyrosinase inhibitory activity in B16 melanoma cells. The active compound in this Cudrania tricuspidata extract was isolated and identified as Flaniostatin by ESI-Mass spectrum, 1H-NMR and 13C-NMR analysis. During our on-going attempts to develop a new skin-whitening agent, we identified a novel candidate compound Flaniostatin. The non-toxicity and tyrosinase inhibitory effects of Flaniostatin are investigated in B16 melanoma cells. In addition, the antioxidant activity of Flaniostatin was evaluated in the oxygen radical absorbance capacity assay, total radical trapping antioxidant potential and DPPH radical scavenging assay. Flaniostatin was shown to be a promising ingredient that could be useful for treating hyperpigmentation and for protecting against oxidative stress.
