Activity of lysosomal enzymes which has antimicrobial and anticancerous functions was altered by some stimuli factor such as oxidative and starvation stress. Unlike lysosomes, peroxisomes have different cellular functions because they contain oxidative enzyme. In this study, the lysosomal activity under the several stresses conditions and characterizing the suitable conditions to separate function of lysosomes and peroxisomes based on subcellular distribution. To certify to organelles’ special function, several chemicals and different concentrations of glucose were used to stimulate in vitro function of lysosomes and peroxisomes isolated from Saccharomyces cerevisiae. The antimcirobial and anticancerous activity of lysosomes was enhanced as a result of subcellular distribution of lysosomes and peroxisomes, and also melanin pigments intensity was reduced. Moreover, we performed a proteomic analysis of lysosomal enzymes in response to the enhanced and repressed conditions for in vitro activity using two-dimensional gel electrophoresis (2-DE). Among analyzed protein, aminopeptidase Y (APE3) was found as the most up-regulated protein in response to oxidative stress. Then, the gene encoding for the APE3 gene was recombinanted and overexpressed in S. cerevisiae in order to confirm its activity in lysosomes. As a result, recombinant S. cerevisiae had antimicrobial effects more than normal cells, besides those were increased more over by galactose induction. We also found that anticancerous effects of lysosomes isolated from recombinant S. cerevisiae were increased against cancer cells such as HeLa cells and melanoma cells, in vitro assay. In addition, bacterial growth in blood was inhibited by lysosomes when we injected lysosomes for therapy agents of bacterimia in rat model. Our results indicated that the increase in endogenous levels of lysosomes and their activity following genetic modification may have various applications as antimicrobial agent and apoptosis-inducing materials for cancer cells. Therefore, it is very promising to use the organelles from genetic modified cells for improving in vitro and in vivo functions and peroxisomal extract as useful agent for skin lightening cosmetics and treatment of hyperpigmentation disorders.
