Among various engineered nanoparticles being used today, nano-scale zinc oxide (N-ZnO) is one of the most widely used nanoparticles in consumer products. Recently, N-ZnO has been used in the food industry as additives and in packaging due to their antimicrobial properties. This increases the likelihood of human exposure to these nanoparticles through oral routes. The present in vivo study in rats was aimed at investigating the repeated oral toxicity of N-ZnO. Therefore, this study is to understand the potential toxicity of N-ZnO through repeated oral toxicity for a period of 28 days. As a contrast group, the micro-scale ZnO (M-ZnO) was studied as well. To understand in vivo accumulation, toxicokinetics and distribution of nanoparticles in target tissues were investigated. Additionally, to understand the mechanism of cell damage, oxidative stress-related markers were measured in the target organs. To evaluate the toxicity of repeated oral administration, N-ZnO and M-ZnO were orally administered to SD rats at 200, 640 and 2000 mg/kg/day for 28 days. During the experimental period, clinical signs, mortality, body weights, food consumption, ophthalmoscopy, urinalysis, hematology, serum biochemistry, gross findings, organ weights and histopathology were examined. The N-ZnO treated groups showed more severe symptoms of lethargy, vomiting and diarrhea on the beginning day after treatment than that of M-ZnO treated groups. Two rats in the N-ZnO 2000 mg/kg treated group were found dead in the 3 week of treatment. Blood hematology test showed that white blood cell, platelets and reticulocytes were significantly increased, and red blood cell (RBC), hemoglobin, hematocrit, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean cell hemoglobin concentration were decreased in both the N-ZnO and M-ZnO 2000 mg/kg treated groups compared to the control. These results indicated that N-ZnO and M-ZnO could cause severe anemia and inflammation. Mild anemia was observed in the N-ZnO 640 mg/kg treated group as well. Furthermore, in the N-ZnO 2000 mg/kg treated group, RBC, hemoglobin, MCV, MCH and hematocrit significantly decreased compared to the M-ZnO 2000 mg/kg treated group, which implied that N-ZnO could induce more severe anemia than M-ZnO. The biochemical liver function tests of serum showed significant elevation of alanine aminotransferase (ALT), aspartate aminotransferase and alkaline phosphatase (ALP), but albumin and total protein decreased in the N-ZnO treated groups. Also, ALT and ALP were increased in the M-ZnO treated groups compared with the control. Biochemical changes in serum indicated that the toxic damage in liver was induced by both N-ZnO and M-ZnO. Such biochemical concentration of serum was generally higher in the N-ZnO treated groups than in the M-ZnO treated groups, which implied that N-ZnO could induce more severe liver damage than M-ZnO. The biochemical renal function tests of serum showed that blood urea nitrogen and creatinine in the N-ZnO treated groups markedly increased either compared with the M-ZnO treated groups or with the control, but no significant difference was found between the M-ZnO treated groups and the control. Furthermore, the renal toxicity of the N-ZnO treatment is confirmed. Severe renal lesions were found by the renal histopathological examination in the N-ZnO treated group. Besides the pathological lesions in the liver, heart, spleen, stomach and intestine tissue were found in all the ZnO treated groups, except N-ZnO 200, M-ZnO 640 and 200 mg/kg treated groups. In the toxicokinetics results, AUC0-24h significantly increased in all the ZnO repeated treated groups for 28 days compared to ZnO single treated groups for 1 day, which indicated that N-ZnO and M-ZnO could cause systemic accumulation. However, AUC0-24h increased 10-fold in the N-ZnO 2000 mg/kg 28 days repeated treatments group and 6.5-fold in the M-ZnO 2000 mg/kg repeated treatments group for 28 days compared to each ZnO 2000 mg/kg single treatment group. Therefore, N-ZnO could induce more severe systemic accumulation than M-ZnO. Also, tissue distribution of N-ZnO 2000 mg/kg treated group showed significantly increased in liver, spleen, kidney and heart compared to M-ZnO 2000 mg/kg treated group and control. reactive oxygen species, malondialdehyde levels in the target organs of N-ZnO 2000 mg/kg treated group significantly increased and superoxide dismutase, glutathione, catalase levels significantly decreased compared to M-ZnO 2000 mg/kg treated group and control. All above results demonstrated that N-ZnO could induce more severe toxic effects than M-ZnO. These enhancement of toxic effects of N-ZnO treated groups may be due to more severe systemic accumulation, oxidative stress mediated cell damage than the M-ZnO treated groups. Additionally, in the 28 days repeated dose toxicity study, no-observed adverse effect level (NOAEL) was below 200 mg/kg/day for N-ZnO and 200 mg/kg/day for M-ZnO, respectively. Also, tissue distribution of N-ZnO 2000 mg/kg treated group showed significantly increased in liver, spleen, kidney and heart compared to M-ZnO 2000 mg/kg treated group and control. reactive oxygen species, malondialdehyde levels in the target organs of N-ZnO 2000 mg/kg treated group significantly increased and superoxide dismutase, glutathione, catalase levels significantly decreased compared to M-ZnO 2000 mg/kg treated group and control. All above results demonstrated that N-ZnO could induce more severe toxic effects than M-ZnO. These enhancement of toxic effects of N-ZnO treated groups may be due to more severe systemic accumulation, oxidative stress mediated cell damage than the M-ZnO treated groups. Additionally, in the 28 days repeated dose toxicity study, no-observed adverse effect level (NOAEL) was below 200 mg/kg/day for N-ZnO and 200 mg/kg/day for M-ZnO, respectively.
