Spraque-Dawley rats were randomly divided into 8 (1 control, 1 vehicle-control, 5 experimental and 3 recovery) groups. Different concentrations of ZnO NPs were dermally applied to the rats in the experimental groups for 90 days. Clinical observations, and weight and food consumptions were measures and recorded daily. Hematologic and biochemical parameters were determined. Gross-pathologic and histopathologic examinations were performed on selected tissues from all animals. Analyses of tissue distributions were done to determine the target organ. There was no mortality change in the experimental group. Although there was a dose-dependent irritation at the site of ZnO application, there was no abnormal finding, which related to ZnO NPs in other organs. The skin penetration of ZnO NPs seemed to be very limited through dermal route. This study demonstrated that the No Observed Adverse Effect Level (NOAEL) of ZnO NPs was above 1000 mg/kg body weight when they were applied dermally.
