Most studies on SV40 large T antigen are focused on binary fissional yeast and animal cells. In such cells, Large T-ag targets several cellular proteins, such as p53 and the Retinoblastoma (Rb)-family of tumor suppressors that control fundamental processes. Recently, it has been reported that the SV40 large T-antigen interacts with specific cellular proteins that cause slowing and even blocking of mitotic progression. The main target for the SV40 large T-antigen is the Rb-family, to manipulate the period of the S-phase via controlling the E2F transcription factor. We have not reported any studies on the effect of the SV40 T-antigen on plant cells, especially on multiple fissional types. This study was conducted to determine whether SV40 T-ag had the same effect on the mitotic division of two multiple fissional microalgal species Scenedesmus quadricauda and Chlorella vulgaris. These two microalgal species are commonly used in biodiesel production. The two species have been chosen to test the possibility of using the SV40 combination in future researches to enhance the expression efficiency of foreign genes. To create three different expression levels, we controlled the expression of the SV40 T antigen using three commonly used heterogenous promoters (NOS, CaMV35S, and CMV). For rapid screening of the effect on cellular mitotic division, the transformed cells were not separated after electroporation. The presence of electroporated vectors was checked once every two weeks by PCR. We observed in generation 0 (G0) of Chlorella cultures that the lowest band concentration was for the vectors containing the T-antigen under the control of the CaMV35S and CMV promoters. The remaining vectors controlled by the NOS promoter were in almost equal concentration compared to the original vector. However, in G1, we could not detect any band for any of the vectors including the empty vector sample which explain there is no vector stability. On the other hand, we observed that in Scenedesmus cultures, band concentrations in G0, G1, and G3 were almost the same compared to the original empty vector. Scenedesmus sp. results indicate no effect on cellular mitotic division because the expression of the T-antigen has been silenced.
