Bioenergy sources may appropriately be regarded as future alternative. Among bioenergy sources, microalgae constitute one of the best alternatives to fossil fuels. Microalgae are photosynthetic microorganisms that are able to rapidly generate biomass and can be used in feasible environmental applications such as wastewater and flue gas treatments. Microalgae can also be utilized for high-value end-products including transport fuels, human and animal feed supplements, industrial chemicals, cosmetics and pharmaceuticals. To utilize microalgae in environmental applications and high value products, selection of potential strain is a most important step, with cell productivity as a key factor. Cell productivity is influenced by factors such as light cycle and carbon source. Light is the major energy source for photosynthetic microorganisms. During the light reaction, light is absorbed by chlorophyll and transformed into ATP and NADPH for reduction of CO2 to organic compounds. Carbon is the major energy source for photosynthetic microorganisms during the dark reaction. Therefore, the light cycle and carbon dioxide bubble size are considered to be important factors in the productivity and yield of photosynthetic reactions. In this study, strains of green micro-algae, Haptophytes and cyanobacteria cells were compared based on chlorophyll a content. Subsequently, lipid content and gene expression were determined. Arthrospira (Spirulina) platensis KMMCC CY-007 showing fast growth and high lipid content, in addition to high level gene expression of photosynthetic components was selected. Following strain selection, the objective of the research presented in this paper was to evaluate the influence of the light:dark period as key growth conditions on the biomass, lipid content and gene expression of Arthrospira platensis KMMCC CY-007. The influence of CO2 bubble size on the cell growth was also evaluated. The effect of light cycle was evaluated at 6:18, 12:12, 16:8, and 24:0 (light:dark periods), respectively. The experiments were conducted at 30 ℃. The air was enriched with carbon dioxide at a concentration of 1 % during continuous aeration at 0.5L/min, and light intensity of 40 μmol m-2 s-1. As the light period increased, morphological length, pH and the amount of biomass were increased. The biomass of the 16:8 light:dark cycle and 24:0 was almost similar. The 16:8 light:dark cycle resulted in the highest yield of lipids 28%, 281.62mg at 7day. The fatty acid methyl ester yields (%) of the lipids accumulated in Arthrospira platensis KMMCC CY-007 showed little differences caused by light:dark cycles. But with 16:8 and 24:0 light:dark cycles, the long chain fatty acid portion was increased, suggesting that potential biodiesel storage stability will be increased. The mRNA level of major photosynthetic enzymes especially PsaC, PsbA and rca were clearly affected by the light period. Interestingly, under 16:8 light;dark cycle higher mRNA expression was detected than for the 12:12 light;dark cycle. Synthetically, 16:8 light;dark cycle is optimal cycle is optimal for biofuel production. However, using the fine bubble, 18:6 light:dark cycle was found to be more profitable for carbon dioxide fixation and biofuel production. The results from this study show that, based on the high lipid quantity of Arthrospira platensis KMMCC CY-007 and CO2 fixation rate, it could serve as a future source of energy feedstock. Also, this study showed optimal conditions of photoperiod and CO2 bubble size based on RNA expression level of Arthrospira platensis KMMCC CY-007.
