유/무기 광감응소재로부터 광여기된 전자에 의한 산화환원 생체촉매의 활성화 및 아밀로이드 펩타이드의 자기조립 제어 현상을 연구하고, 이를 인공광합성 기반 고부가가치 화학물질/연료 생산 및 아밀로이드 응집억제를 통한 알츠하이머 동물모델의 광역학적 치료에 응용하는 것을 최종목표로 한다.

유/무기 광감응소재로부터 광여기된 전자에 의한 산화환원 생체촉매의 활성화 및 아밀로이드 펩타이드의 자기조립 제어 현상을 연구하고, 이를 인공광합성 기반 고부가가치 화학물질/연료 생산 및 아밀로이드 응집억제를 통한 알츠하이머 동물모델의 광역학적 치료에 응용하는 것을 최종목표로 한다.
연구개발내용
1단계 연구에서는 생체촉매 활성화를 위한 유/무기 광감응소재 개발을 통해 효율적인 인공광합성 시스템을 구축하였다. 자연계 광합성의 Z-scheme을 모방한 광전기화학적 셀을 구성하여 인가되는 외부전압을 낮춘 효율적인 광여기 전자 전달 시스템을 개발하였다. 구체적으로 물 분해 촉매(Co-Pi)가 고정화된 hematite 광양극과 BiFeO3 광음극으로 이루어진 광전기화학셀을 완성하였고, 광여기된 전자를 통해 재생된 조효소는 3-효소 연속반응 시스템과 연계되어 이산화탄소를 메탄올로 성공적으로 전환하였다. 또한 rose bengal을 광감응제로 사용하여 ene-reductase 생체촉매의 prosthetic group에 직접적인 전자전달을 유도하여, 값비싼 조효소(NADH)의 재생과정없이 훨씬 간단한 방법으로 효소 반응을 촉매하는 인공광합성 시스템을 개발하였다. 지속적 인공광합성 시스템을 구축하기 위해선 광흡수체, 전자 매개체 및 산화환원촉매와 같은 주요 광합성 구성요소가 기능 활성에 영향 없이 계층적 구조로 캡슐화 됨이 필요하다. 이에 따라 집광형 자기조립 펩타이드 하이드로젤 소자를 개발하고 하이드로젤 구조체 안에서 광화학적 조효소 재생과 산화환원 효소 반응이 효율적인 물질전달을 통해 일어나도록 구축하였다. 또한 광감응성 물질의 광여기 현상에 착안, 광감응제 및 광전극소재와 빛 조사를 통한 아밀로이드 펩타이드의 자기조립 억제 작용을 분석하였다. 구체적으로 메틸렌 블루 염료와 탄소 기반의 카본 나노닷 및 산화철 광전극을 광감응제로써 사용하였으며, 특히 메틸렌 블루의 경우, 알츠하이머 초파리 모델에서 광감응 작용을 통해 생체 내 아밀로이드 펩타이드의 응집을 효과적으로 억제한다는 것이 입증되었다. 카본 나노닷은 탄소 원료로 합성되기 때문에 생체적 합성이 뛰어나고 높은 수용성 및 광학적 특성을 가져 생체용 광감응제로 적합한 물질이다.
이를 더 높은 광학적 안정성과 가시광선 영역대의 반응성을 가지도록 표면개질한 뒤 아밀로이드 펩타이드와 혼용하여 아밀로이드 펩타이드의 자기조립 양상을 어떻게 변화시키는지 살펴보았다. 본 연구의 메틸렌 블루와 카본 나노닷을 이용한 광감응 펩타이드 자기조립 제어기작은 펩타이드 응집 억제뿐만 아니라, 이미 형성된 독성의 응집체를 분해하고 체내 독성을 줄일 수 있어 아밀로이드 표적의 알츠하이머병 예방과 치료에 대한 잠재성을 보여준다.
뇌 직접 삽입을 통한 응집 억제 방법인 전극 소재 활용의 경우, 가시광선 영역에서 반응하는 에너지 준위를 가지고 화학적으로 안정한 산화철이 이용되었다. 양전압과 빛 조사를 가하여 산화철의 전자가 여기될 수 있는 환경이 조성되면 광전극으로부터 발생되는 수산기 라디칼이 아밀로이드의 응집을 억제함을 확인하였고, 전극 표면에 촉매가 도입될 경우 그 억제 효과가 강화되는 기작을 성공적으로 유도함으로써 펩타이드 자기조립이 빛 조사 및 실험조건에 따라 제어될 수 있는 시스템을 개발하였다.

인공광합성기술은 에너지원으로 무한한 태양광을 사용하여 화학연료, 정밀화학제품 등을 생산한다는 장점 때문에 그 파급효과가 매우 크며, 본 연구에서 제안하는 빛을 이용한 보조인자 비의존적 생체촉매기반 인공광합성 시스템을 이용한 의약품 개발 및 다양한 바이오연료의 생성에 관한 연구는 아직 시스템 구축이 이루어진 바가 없는 독창적인 연구임. 또한 초고령사회 진입을 앞두고 있는 현재, 인간의 10대 사망원인 질환 중 유일하게 알츠하이머병만이 아직까지 예방, 진단, 및 치료법이 전무한 상태이며, 본 창의과제에서 제안하는 빛을 이용한 알츠하이머 아밀로이드 응집억제를 이용한 광역학적 치료기법은 세계적으로도 시도된 바가 없는 연구임.

The CRI project aims at the design of photosensitizing platforms for visible light-induced, water-oxidation-driven activation of redox biocatalysts and the photodynamic control of amyloid peptide self-assembly towards solar chemical synthesis by biocatalytic artificial photosynthesis and photosensitized inhibition of amyloid aggregation and toxicity in vitro and in vivo.

In the 1st stage of this project, we have developed artificial photosynthetic systems through synthesis of organic/inorganic photosensitive materials. To take advantage of the natural photosynthetic Z-scheme, we designed a Co-Pi/hematite|BiFeO3 tandem photoelectrochemical (PEC) cell for NADH regeneration by proper alignment of the band edges of two photoelectrodes. The tandem PEC cell was successfully integrated with a multienzyme cascade system for the biocatalytic reduction of CO2 into methanol. We have applied rose bengal as a photosensitizer to activate ene-reductases through direct photoinduced electron transfer, while eliminating the necessity for the supply of expensive NADH cofactors and its generation. For the implementation of sustainable artificial photosynthesis, key photosynthetic components need to be encapsulated into a hierarchical architecture without the loss of their activities. As a result, we developed self-assembled, light-harvesting peptide hydrogels encapsulating electron mediators, cofactors and biocatalysts for biomimetic photosynthesis. Based on the excitation of photosensitizers, we have explored the mechanism of suppressing amyloid aggregation in the presence of light and photoactive materials. We have applied photosensitizing materials such as Methylene blue, carbon nanodots and hematite.
In particular, we validated in-vivo inhibitory efficacy of Methylene blue against amyloid aggregation by using Dorsophila model of Alzheimer’s disease. With the merits of carbon nanodots such as high biocompatibility, hydrophilicity and optical stability, we further functionalized the surface of carbon nanodots to enhance photoactivity and analyzed the effect of resulting carbon nanodots on amyloid aggregation. In this project, we have verified the effects of Methylene blue and carbon nanodots in inhibition of amyloid aggregation and disaggregation of amyloid aggregates, which show the great potential in amyloid-targeted Alzheimer’s disease treatment. In the case of using hematite as a photoelectrode, photoexcited electrons generated hydroxyl radicals under the positive voltage and light, inducing the suppression of amyloid aggregation. Furthermore, by doping the catalyst onto photoelectrode, we have succeeded the enhancement in suppressing efficacy of hematite and the modulation of amyloid self-assembly characteristics.

Expected Contribution
This CRI project will have significant impact on both energy and healthcare fields. This project will explore how to unite those two catalytic functionalities into a creative breakthrough of biocatalytic artificial photosynthesis for the ultimate goal of utilizing solar energy. Biocatalytic artificial photosynthesis will create a technological foundation for solar synthesis of fine chemicals and fuels. From the healthcare aspect, the proposed research will provide new insights toward photodynamic therapy of Alzheimer's disease in future by studying the effect of different photosensitizing materials against amyloid aggregation and toxicity.
