전통한약처방인 당귀작약산은 예로부터 혈행 부족으로 인한 노인성 질환의 치료에 사용되어 왔다. 최근 허혈성 뇌손상에 대한 신경세포재생성 치료법이 많은 학자들에게 주목받고 있다. 본 연구에서는 당귀작약산이 전뇌허혈 모델에서 뇌 해마지역의 신경 세포재생성 효과를 발휘하는지와 그 기전에 대해서 조사하였다. 먼저 수동회피시험에서 당귀작약산이 정상 동물의 학습 및 기억에 미치는 효과와 세포 분열에 미치는 효과를 알아보았다. 다음으로 이러한 효과를 바탕으로 당귀작약산의 전뇌허혈 후투여가 전뇌허혈로 유도되는 기억력 감퇴와 신경세포신생, Akt/GSK3β/β-catenin 신호전달에 미치는 효과를 알아보았다. 당귀작약산을 정상 mice에게50, 100, 200 및 400 mg/kg의 농도로 14일간 경구로 투여한 결과, 수동회피시험에서 100 및 200 mg/kg 의 농도에서 학습 및 기억력을 향상시키며 세포 분열 정도를 증가시킴을 확인할 수 있었다. 그리고 전뇌허혈 후 당귀작약산의 4주간 투여는 모리스 수미로 시험에서 뇌허혈로 유도되는 공간 기억력의 감퇴를 개선시킴을 확인하였다. 또한 당귀작약산이 뇌허혈로 유도되는 신경세포신생을 촉진시키며 Akt/GSK3β/β-catenin 신호전달을 증가시킴을 알 수 있었다. 이러한 결과는 당귀작약산이 뇌허혈로 유도되는 신경세포 사멸 및 그로 인한 뇌기능 손상에 대한 치료약물로서의 개발 가능성을 제시한다.

Recently, regenerative medication for the treatment of cerebral ischemia has drawn the attention of many researchers. In this study, it was examined whether DJS exerts a neuronal regenerative effect in the hippocampus of a transient forebrain ischemic mice model. To clarify this, the effect of DJS (50, 100, 200, and 400 mg/kg/day, p.o., for 14 days) on passive avoidance memory and neural proliferation in normal mouse brain were examined. Next, using this result, the effect of delayed administration of DJS (100 mg/kg/day, p.o.) on transient forebrain ischemia-induced memory impairment, neurogenesis, and Akt/GSK3β/β-catenin signaling were tested. It was observed that DJS (100 or 200 mg/kg/day, for 14 days) administration enhanced passive avoidance memory and neural proliferation. Moreover, the delayed administration of DJS (100 mg/kg/day, p.o.) ameliorated ischemia-induced spatial memory impairment in the Morris water maze task. DJS (100 mg/kg/day, p.o.) facilitated ischemia-induced neurogenesis. Akt/GSK3β/β-catenin signaling was also increased by DJS treatment. These findings confirm the effect of DJS on insufficient blood supplies-induced gerontological disorders. Moreover, it could be suggested that DJS is a possible candidate for the treatment of ischemia-induced neuronal degeneration.
