Daidzein (1), formononetin (2), genistein (9)의 염증 억제 효과의 기전을 알아보고자 HO-1 발현을 확인하였으나, HO-1은 발현되지 않았으며, HO 활성도 증가하지 않았다. 염증반응에서 주요한 기전으로 알려진 NF-κB 활성에 대한 실험에서는 daidzein (1), formononetin (2), genistein (9) 모두 유의하게 억제하는 것을 확인하였다.

Pharmacological investigations and clinical practices have shown that isoflavones are one of the most important contributors to the biological activity of Puerariae Radix, including the prevention of cardiovascular diseases and cancers, anti-diabetic and anti-oxidative activities, neuroprotective effects and estrogenic activity. Among the Puerariae Radix isoflavones, puerarin is believed to be the most abundant. Recent investigations have demonstrated that puerarin shows anti-oxidant, anti-hypercholesterolemia, antiplatelet aggregation and anti-inflammatory activities. Glutamate-induced oxidative injury contributes to neuronal degeneration in many central nervous system (CNS) diseases, such as epilepsy and ischemia. In the present study, I investigated whether the compounds isolated from Puerariae Radix, possessed neuronal cytoprotective effects against glutamate-induced oxidative injury in mouse hippocampal HT22 cells. The bioassay-guided fractionation of the ethylacetate soluble part of 50% EtOH extracts from the Puerariae Radix led to the isolation of eleven compounds. Daidzein (1), formononetin (2), daidzin (3), puerarin (4), genistin (5), derriscandenoside A (6), 4’,8-dimethoxyisoflavone-7-O-β-D-glucose (7), 8-O-methylretusin (8), genistein (9), coumestrol (10) and lupeol (11) were isolated from the ethylacetate soluble part of 50% EtOH extracts from the Puerariae Radix. 8-O-methylretusin (8) and coumestrol (10) showed the potent neuroprotective effects on glutamate-induced neurotoxicity and induced the expression of heme oxygenase (HO)-1 in the mouse hippocampal HT22 cells. Microglial activation has been implicated in various neurodegenerative diseases such as Alzheimer’s disease and Parkinson’s disease. In this study, isoflavonoids isolated from Puerariae Radix were examined to see their abilities to inhibit pro-inflammatory factors in lipopolysaccharide (LPS)-stimulated murine BV2 microglia cells. Results showed that daidzein, genistein and formononetin significantly inhibited LPS-induced pro-inflammatory mediators, such as nitric oxide (NO), prostaglandin E2 (PGE2), and tumor necrosis factor-α (TNF-α), without any significant cytotoxicity. On the other hand, puerarin, daidzin and genistin failed to suppress such mediators. Moreover, daidzein, genistein and formononetin also downregulated protein expression of inducible NO synthase (iNOS), and cyclooxygenase-2 (COX-2) in LPS-induced BV2 microglia cells, while expression of iNOS and COX-2 were not changed in cells treated with puerarin, daidzin and genistin. Furthermore, it was also revealed that daidzein, genistein and formononetin treatment significantly reduced translocation of nuclear factor-κB (NF-κB) subunit p65 and its DNA-binding activity in LPS-stimulated BV2 microglia cells. Taken together, data suggest that 8-O-methylretusin and coumestrol isolated from Puerariae Radix could be the potential therapeutic candidates for ROS-related neurological diseases. In addition, isoflavonoids such as daidzein, genistein and formononetin may play a pivotal role to inhibition of imflammation, providing important implications for therapeutic intervention of neuroinflammation.
